摘  要:目的:探讨线粒体功能在支气管哮喘患者中的变化特点。方法:哮喘组选取2016年4月至2017年5月新疆医科大学附属中医医院门诊及住院确诊为支气管哮喘急性发作期并符合纳入排除标准的患者,共122例,按照病情严重程度分级标准分为4组,间歇状态(第1级) 45例,轻度哮喘(第2级) 23例,中度哮喘(第3级) 30例,重度哮喘(第4级) 24例;正常对照人群来自同期我院体检中心体检的健康正常人,共44例。采血分离血小板及血小板线粒体,透射电镜观察血小板超微结构,检测血小板线粒体膜电位(MMP)、细胞色素C氧化酶(COX)活性、三磷酸腺苷(ATP),分析它们的差异性。结果:哮喘组患者与正常组患者血小板线粒体膜电位(MMP)、细胞色素C氧化酶(COX)活性、三磷酸腺苷(ATP)比较有统计学差异(P<0.05),哮喘组低于正常组;不同病情严重程度的哮喘患者与正常对照组的线粒体功能(MMP、ATP、COX)有统计学差异(P<0.05)。结论:哮喘患者机体内存在着线粒体呼吸链的抑制和功能的受损,线粒体损伤和功能障碍可能对哮喘发病有重要影响。Objective: To investigate the changes of platelet mitochondrial function in asthmatic patients.Methods: 122 cases were diagnosed as acute episode of asthma from April 2016 to May 2017. According tothe severity grading standard, they were divided into 4 groups, 45 cases of intermittent state (first grade), 23 cases of mild asthma (second grade), 30 cases of moderate asthma (third grade) and 24 cases of severe asthma (fourth grade). 44 cases of normal control group came from the physical examination center at same time.Platelet and platelet mitochondria were separated from blood samples. The ultrastructure of platelets was observed by transmission electron microscopy. The platelet mitochondrial membrane potential (MMP ), cytochrome C oxidase (COX) activity and adenosine triphosphate (ATP) activity were detected and their differences were analyzed. Results: There were significant differences in platelet mitochondrial membrane potential(MMP), cytochrome C oxidase (COX) activity and adenosine triphosphate (ATP) between asthma groupand normal group (P〈0.05). The mitochondrial function (MMP, ATP, COX) of asthmatic patients with different severity was significantly different from that of normal control group (P〈0.05). Conclusion: There aremitochondrial respiratory chain inhibition and impaired function in asthmatic patients. Mitochondrial damageand dysfunction may play an important role in the pathogenesis of asthma.
