摘 要:[目的]为了明确番茄病程相关基因在病原菌胁迫下的表达模式,[方法]本研究运用实时荧光定量PCR法对番茄中PRP5、PRP10、DRRP206、ERTF1B和PO等基因在尖孢镰孢菌番茄专化型菌株胁迫条件下和非胁迫条件下3 h、6 h、12 h、24 h、48 h和7 d时间段的表达情况进行研究。[结果]结果表明,供试的番茄植株在接种7天后表现出明显的萎蔫症状;PRP10基因在处理组中的相对表达量远低于同一时间段对照组中的相对表达量;ERTF1B基因在处理组和对照组中各时间段相对表达量值极低,差异不明显;在处理组中PRP5、DRRP206和PO基因在接种7 d后的相对表达量明显高于对照组,其中DRRP206和PO基因在接种12 h后的相对表达量显著上升。[结论]综上所述,在病原菌胁迫下番茄感病品种中病程相关蛋白编码基因PRP5、PRP10和ERTF1B的相对表达量较低甚至不表达,还有些病程相关蛋白编码基因如DRRP206和PO的相对表达量较高,但并不足以使供试的番茄植株抗病。[Objective]Present study was proposed to clarify the expression patterns of pathogenesis related genes in to mato under the stress of pathogen infection. [Methods] Five pathogenesis related genes, PRPS, PRP10, DRRP206, ERTF1B, and PO, were selected, and extracted at 3, 6, 12, 24, 48 h, and 7 d after inoculating with F. oxysporum f. sp. Lycopersici. The expression patterns of selected genesin tomato seedlings were analyzed byRT qPCR. The seedlings treated with sterilized water were served as Control treatment. [Results] The results showed that tomato plants displayed obvious wilt symptom at 7 days after inoculation. The relative expression level of PRPIO gene in the treatment wassignificantly lower than that in the control treatment, and ERTF1Bgene in both treated and control groups expressed extremely low level, and no statistic differences were observed between treated and control groups. At 7 d post treatment, the relative expression levels of PRP5, DRRP206and PO in the treated group were significantly higher than those in the control group, furthermore, the relative expression levels of DRRD206 and PO in the trea ted group were significantly increased at 12 h post treatment. [Conclusion]In summary, under the stress of pathogen infection, the relative expression level of pathogenesis related genes PRPS, PRPIO, and F.RTF1B in tomato suscepti ble varieties were fairlylow or non expressed, while DRRP206 and PO gene were highly expressed, however, they were notsufficient to endow the disease resistance to the tested plant.
