摘 要:[目的]发掘脂肪酸代谢关键酶基因,了解其编码蛋白的理化性质,探究该基因在紫苏不同组织中及不同逆境胁迫下的表达特性,为紫苏分子育种提供基因元件。[方法]利用生物信息学技术对转录组测序数据库中获得的紫苏PfPDAT1基因序列进行了分析,并采用qRT-PCR技术研究了不同逆境胁迫下该基因的表达特性。[结果]PfPDAT1基因cDNA全长序列为2 097 bp,包含1 554 bp的开放阅读框,编码517个氨基酸残基,通过预测将PfPDAT1基因编码蛋白定位于细胞质;多序列比对分析显示,PfPDAT1基因编码蛋白与芝麻和油橄榄具有较高的同源性,序列相似性分别为89%和83%;实时荧光定量PCR分析结果表明,PfPDAT1基因在‘晋苏1号’不同组织中均有表达,且在种子中表达量最高;PfPDAT1基因在NaCl、PEG6000和低温胁迫诱导时,其相对表达量有显著差异,且均在胁迫6 h时表达量达到最高。[结论]PfPDAT1基因在紫苏生长发育及抵御逆境胁迫过程中起重要作用。[Objective]Present study was conducted to explore key enzymatic genes which regulate fatty acid metabolism, clarify the physical and chemical characteristics of their encoded proteins, and determine the expression characteristics of PfPDAT1 in different organs of Perilla under different stresses, and aimed to provide solid information for Perilla molecular breeding. [Methods] In this study, the PfPDAT1 gene sequence obtained from the transcriptomic database was analyzed using bioinformatic methods, and the expression characteristics of this gene under different stresses were studied by qRT PCR. [Results] The full length cD02222NA sequence of PfPDAT1 gene of Perilla was 2097 bp, and it contained 1554 bp open reading frame (ORF) which encoded 517 amino acid residues. The PfPDAT1 protein was predicted to be located on the cytoplasm. Multiple sequence alignments analysis revealed that PfPDAT1 had close ge netic relationship with its homologs of Sesamum indicum and Olea europaea, and the sequence similarity was up to 89 and 83%, respectively, qRT PCR result showed that PfPDAT1 gene was expressed in all organs of 'Jinsu 1', and the highest expression level of PfPDAT1 was observed in seeds. In addition, the expression of PfPDAT1 was signifi cantly induced by NaCl, drought, and cold stress, and the expression level was peaked at 6 h. [Conclusion] The PfP DAT1 gene plays an important role in growth and development of Perilla.
