摘 要:目的:研究3'-大豆苷元磺酸钠对大鼠脑缺血/再灌注损伤后金属蛋白酶-9保护作用及其机制。方法:大鼠大脑中动脉栓塞制作局灶性脑缺血/再灌注损伤模型,缺血后10 min,于舌下静脉注射3'-大豆苷元磺酸钠(2. 0 mg·kg^(-1))。再灌注24 h后。取缺血区脑组织制成10%的匀浆,定量PCR检测法测定MMP-9,Gfap,Dlg4,Chrna7 mRNA的表达。结果:与模型组相比,3'-大豆苷元磺酸钠能显著降低MMP-9,GfapmRNA的表达,能显著升高Dlg4和Chrna7 mRNA的表达。结论:3'-大豆苷元磺酸钠对大鼠脑缺血/再灌注损伤的保护作用机制,可能与其降低MMP-9、Gfap mRNA的表达和升高Dlg4、Chrna7 mRNA的表达有关。Objective: To investigate the protective effects of 3′-daidzein sulfonate sodium (DSS) with MMP-9 on occludin following cerebral ischemia reperfusion injury (CIRI) in rats, and the mechanism of these effects. Methods: The rat CIRI model was established by middle cerebral artery occlusion, and received sublingual venous injection of 3′-DSS (2.0 mg·kg^ -1 ) 10 min after ischemia. 24 h after reperfusion, ischemic cerebral tissues were collected and prepared into 10% homogenate, which was used for quantitative PCR assay of the expression of MMP-9,Gfap,Dlg4,Chrna7 mRNA. Results: Compared to the model group, 3′-DSS significantly reduced expression of MMP-9,GfapmRNA, and significantly increased expression of Dlg and Chrna7 mRNA. Conclusion: The protective effects of 3′-DSS on CIRI in rats might be associated with its potential to reduce expression of MMP-9、Gfap mRNA and to increase expression of Dlg4、Chrna7 mRNA.
