摘 要:目的:观察辛通畅络法肾络宁对膜性肾病(MN)模型小鼠基质金属蛋白酶-9 (MMP-9)、白介素-10 (IL-10)的影响。方法:实验Balb/c小鼠采用阳离子化牛血清白蛋白(C-BSA)诱导膜性肾病模型,分为模型组、肾络宁组和MMP-9基因敲除组,另设空白对照组,其中基因敲除组于造模前行MMP-9基因敲除。实验以肾络宁为治疗药物,于实验第0周、第6周末检测各组小鼠尿蛋白,血清IL-10、MMP-9的变化情况。结果:治疗6周后,模型组和肾络宁组、基因敲除组小鼠均出现不同程度的尿蛋白排泄量增加,其中肾络宁组、基因敲除组较模型组小鼠尿蛋白排泄量减少;MMP-9基因敲除组小鼠MMP-9表达显著降低,肾络宁组和模型组有不同程度升高,其中肾络宁组MMP-9表达较模型组降低;模型组、肾络宁组、基因敲除组IL-10表达均匀不同程度降低,肾络宁组、基因敲除组高于模型组(P﹤0.05)。结论:肾络宁对MN模型小鼠具有减少尿蛋白排泄的作用,其机制可能与调节IL-10及MMP-9的表达相关。Objective:To observe the effect of Shenluoning on matrix metalloproteinase-9 (MMP-9)and interleukin-10 (IL-10) in model mice with membranous nephropathy (MN). Methods:The experimental Balb/c mice were induced as MN model by cationic bovine serum albumin (C-BSA),and divided into model group,Shenluoning group and MMP-9 gene knock-out group. The blank control group was established in addition. Among them,the gene knock-out group was knocked-out MMP-9 gene before modeling.In the experiment,Shenluoning was taken as therapeutic drug. On the 0th week and 6th weekends of experiment,the changes of urine protein,blood-serum IL-10 andMMP-9 of mice in each group were detected. Results:After 6 weeks of treatment,the mice in model group,Shenluoning group and gene knock-out group increased the excretion of urinary protein in different degrees;among them,the excretion of urinary protein of mice in Shenluoning group and gene knock-out group was lower than that in model group. MMP-9 expression of mice in MMP-9 gene knock-out group was significantly reduced,while MMP-9 expression of mice in model group and Shenluoning group was increased in different degrees;among them,MMP-9 expression of mice in Shenluoning group was lower than that in model group. IL-10 expressions in model group,Shenluoning group and MMP-9 gene knock-out group were lowered in different degrees;Shenluoning group and gene knock-out group were higher than model group (P〈0.05). Conclusion:Shenluoning has the effect of reducing excretion of urinary protein on MN model mice.Its mechanism may be related to regulating the expressions of IL-10 and MMP-9.
