摘  要:目的 探讨姜黄素预处理对干热环境热射病大鼠肺损伤及氧化应激指标变化的影响。 方法 将50只SD大鼠随机分为五组(n=10):常温对照组(NC组)、干热对照组(DHC组)、姜黄素低浓度组(姜黄素50 mg/kg预处理组)、姜黄素中浓度组(姜黄素100 mg/kg预处理组)、姜黄素高浓度组(姜黄素200 mg/kg预处理组)。NC、DHC组给予生理盐水灌胃,姜黄素预处理组给予不同浓度的姜黄素灌胃(1次/d,连续7 d)。第8天除NC组外,其余四组大鼠转移至西北特殊环境人工实验舱内进行实验,环境温度(41±0.5)℃,湿度(10±1)%。实验的第150 min达到热射病状态,麻醉后肺组织取材。肺组织进行HE染色及肺损伤病理学评分,并采用比色法测定肺组织中丙二醛(MDA)、髓过氧化物酶(MPO)、过氧化氢酶(CAT)及超氧化物歧化酶(SOD)。 结果 DHC组肺损伤病理学评分明显高于NC组(P〈0.01),随着姜黄素浓度增加,肺病理学评分呈下降趋势,不同浓度姜黄素预处理组之间差异均具有统计学意义(P〈0.01),DHC组和姜黄素各组较NC组MDA、MPO含量升高,CAT、SOD活力降低;DHC组MDA(96.25±1.34 vs. 73.71±1.12、54.13±1.60、40.531.06)、MPO(6.09±0.16 vs. 5.33±0.17、4.30±0.13、3.75±0.16)含量明显高于姜黄素预处理组(P〈0.01),CAT(2.49±0.06 vs. 2.67±0.04、2.85±1.00、3.17±0.09)、SOD(18.23±3.90 vs. 31.83±4.92、44.57±5.65、55.00±5.03)活力明显低于姜黄素各组(P〈0.01)。Pearson相关分析结果提示,肺病理损伤评分与MDA、MPO之间存在正相关性(相关系数分别为 r=0.945,r=0.962,P〈0.01),与SOD、CAT之间存在负相关性(相关系数分别为r=-0.903,r=-0.935,P〈0.01)。 结论 姜黄素预处理对沙漠干热环境热射病大鼠的肺损伤具有保护作用,姜黄素可能通过调控氧化应激反应来发挥肺损伤保护作用。Objective To investigate the changes of the lung tissue damage and oxidative stress in heatstroke rats under dry heat environment of desert by curcumin pretreatment. Methods 50 Sprague - Dawley rats were randomly divided into five groups (n = 10) : Normal control group (NC group), Dry heat control group ( DHC group), Low concentration group (50 mg/kg curcumin pretreatment group),Middle concentration group (100 mg/kg curcumin pretreatment group), High concentration group (200 mg/kg cureumin pretreatment group). The NC and DHC groups were given a gavage of normal saline, while curcumin pretreatment groups were given a gavage of curcumin with the concentration 50 mg/kg, 100 mg/kg, 200 mg/kg, once a day for 7 consecutive days. At 8th day, all except for NC group were transferred to the climate cabin (The Simulated Climate Cabin for Special Environment of Northwest of China) with the conditions of (41± 0.5 )℃, (10 ± 1 )% relative humidity. At the time of 150th minute since the experiment began, the rats were in heat stroke states, and same done to Control group. Lung tissues were harvested and pathological changes were observed by HE staining. Colorimetric determination of MDA, MPO, SOD and CAT levels. Results Lung injury pathological scores were higher in DHC group than in NC group, the lung injury pathological scores gradually decreased with the increased curcumin concentration, and each group had significant different (P 〈 0.01 ). The content of MDA and MPO in the groups of DHC and cureumin was higher than NC group, and the activity of CAT and SOD decreased. The content of MDA (73.71 ±1.12, 54. 13 ± 1.60, 40.53 ± 1.06) and MPO (5.33 ±0.17, 4.30 ±0. 13, 3.75 ±0. 16) in DHC group was significantly increased than those in curcumin groups ( P 〈 0.01 ), and the activity of CAT (2.67 ±0.04, 2.85 ± 1. 00, 3.17 ±0.09) and SOD (31.83 ± 4.92, 44.57 ± 5.65, 55.00 ±5.03 ) was significantly decreased ( P 〈 0.01 ).
