穴区与非经非穴区胶原纤维含量的研究

摘　　要：目的:研究大鼠后肢太阴脾经"三阴交""阴陵泉""血海"穴区与非经非穴区深面显微形态及胶原纤维的含量。方法:选取10只清洁级SD大鼠,分别对三阴交、阴陵泉、血海进行穴区体表定位。水合氯醛腹腔注射将大鼠麻醉,用10%中性多聚甲醛经升主动脉进行灌注固定直至大鼠四肢僵硬后选取穴区深部组织及非经非穴区深部组织,将深部组织置于4%甲醛溶液中固定。常规脱水透明石蜡包埋后切片,HE染色及Masson三色改良染色,镜下观察穴区深部组织结构、胶原纤维形态与量的变化。结果:"三阴交""阴陵泉""血海"穴区HE染色观察后穴区深部形态学无明显差异,Masson三色改良染色发现穴区结缔组织较非穴区结缔组织所占比值较大(P<0. 01),三阴交穴区的结缔组织百分比含量比阴陵泉和血海区的结缔组织含量大(P<0. 01)。结论:大鼠后肢太阴脾经"三阴交""阴陵泉""血海"穴区有神经血管束或富含神经血管的结缔组织穿行,且三阴交穴区周围筋膜结缔组织含量较阴陵泉、血海穴区更为丰厚。Objective：To study the deep surface morphology and collagen fiber content in both acupoint area as Sanyinjiao, Yinlingquan, Xuehai and non-meridian -non-acupointarea in the posterior limbs of rats. Meth- ods ： 10 clean SD rats were selected and point surface location of Sanyinjiao, Yinlingquan and Xuehai were car- fled out respectively. Rats were anesthetized with intraperitoneal injection of chloral hydrate. 10% neutral poly-formaldehyde was perfused via ascending aorta until rats＇ limbs become stiff. The deep tissues in the acupoint ar- ea and non-meridian-non-acupointarea were selected and fixed in a 4%＇ formaldehyde solution. After conven- tionaldehydration and transparent embedding, sliced, HE stained and modified Masson＇s trichrome stai- ning were performed. The texture of the deep acupoint area and the change of the morphology and the quantity of the collagen fiber were observed under microscope. There was no significant difference in the morphol- ogy in the deep area of the Sanyinjiao, Yinlingquan and Xuehai after HE stained. Modified Masson＇s trichrome staining showed that the ratio of connective tissues in the acupoint area was larger than that in the non-meridian -non-acupeint area （P〈0.01）. The percentage content of connective tissues in Sanyinjiao area was higher than that in Yinlingquan and Xuehai area （P〈0.01）. Conehtsion： There are bundles of nerve vessels or connective tissues rich in nerve vessel in the acupoint area of Sanyinjiao, Yinlingquan and Xuexai in the posterior limbs of rats. The content of fascial connective tissues around Sanyinjiao is more abundant than that around Yinlingquan and Xuehai.
