摘　　要：目的探讨温热疗法对孢子丝菌病模型小鼠皮损局部免疫微环境的影响。方法选取50只BALB/C雌性小鼠,建立孢子丝菌病小鼠模型。将小鼠随机分入A组和B组,每组25只。B组在接种2周后应用恒温水浴箱对小鼠尾部皮损局部进行水浴温热治疗,温度为43℃,每次15 min,每周3次,共治疗4周;A组不予任何处置。比较两组疗效差异,免疫组化法检测皮损病变组织中干扰素-γ的表达水平,聚合酶链式反应法检测皮损病变组织中CD80、CD86的表达水平。结果 B组小鼠皮损消退快于A组。B组小鼠皮损消退率高于A组,差异有统计学意义(P <0. 05)。治疗2周后,B组小鼠皮损病变部位干扰素-γ、CD80、CD86的表达水平均高于A组,差异有统计学意义(P <0. 05)。结论温热疗法治疗孢子丝菌病效果显著,其作用机制可能与皮损病变组织中干扰素-γ、CD80、CD86的表达水平上升有关。Objective To investigate the effect of thermotherapy on sporotrichosis model mice and the influence on local immune microenvironment of skin lesions.Methods A total of 50 female BALB/C mice were selected to establish mice model of sporotrichosis.The mice were randomly divided into Group A and Group B,with 25 mice in each group.Two weeks after inoculation,mice of Group B were applied the thermostatic water bath box to treat the local skin lesions in the tail,the temperature was 43℃,15 minutes each time,3 times a week,totally 4 weeks.The mice in Group A were not performed any treatment.To compare the differences in efficacy between the two groups,immunohistochemistry was used to detect the expression level of interferon-γ（IFN-γ） in skin lesions,and PCR was used to detect the expression levels of CD80 and CD86 in the skin lesions.Results The skin lesions in Group B subsided faster than that in Group A.The regression rate of skin loss in Group B was significantly higher than that in Group A（ P 〈0.05）.After 2 weeks of local hyperthermia treatment,the expression of IFN-γ,CD80 and CD86 were higher than those in Group A（ P 〈0.05）.Conclusion The effect of thermotherapy on sporotrichosis is remarkable,and its mechanism may be related to the increased expression levels of IFN-γ,CD80 and CD86 in the skin lesion tissues.
