摘 要:建立了长春西汀原料和制剂中4种异构体的液相色谱分析方法。采用CHIRALPAK IG-3手性色谱柱,用主成分自身对照法计算异构体质量。结果表明,4种长春西汀异构体色谱峰分离良好,分离度>1. 5,专属性和耐用性强,4种长春西汀异构体色谱峰面积与溶液质量浓度线性关系良好,检出限分别为0. 31、0. 31、0. 45、0. 45 ng;定量限分别为1. 04、0. 78、1. 49、1. 50 ng,相对标准偏差分别为2. 53%、1. 81%、1. 82%、2. 03%(n=6),异构体E、F、G样品加标回收率分别为99. 9%、101. 6%、102. 0%。该方法操作简单、通用性强、测定结果准确,可用于长春西汀原料及制剂的异构体控制。A reverse-phase HPLC method is developed for determining four isomers of vinpocetine and its raw materials. CHIRALPAK IG-3 column is adopted and the isomer contents are calculated by self-contrast method. It is shown that this method can separate four isomers well,with resolution(R)〉1. 5,which has good specificity and durability.The peak areas for four vinpocetine isomers all have good linear relationships with the content in solution.The detection limits for vinpocetine,isomer E,isomer F,isomer G are 0. 31,0. 31,0. 45 and 0. 45 ng,respectively,the quantitation limits are 1. 04,0. 78,1. 49 and 1. 50 ng,respectively,the recovery rates are 99. 9%,101. 6%,102. 0%respectively and the RSD are 2. 53%,1. 81%,1. 82% and 2. 03%(n = 6). This method is simple,accuracy and convenient,and can be reliable for the quality control of isomers of vinpocetine and its preparation.
