摘 要:由于喜树碱内酯环开环后在254 nm波长下紫外吸收增强,而开环的喜树碱毒副作用大。鉴于目前喜树碱含量测定均采用254 nm,且喜树碱在大多数溶剂中溶解性较差,为更准确便捷地检测喜树碱原料药的含量,有必要探索在新的波长下测定喜树碱含量的方法。将喜树碱在200~400 nm扫描,发现在367nm处有较大吸收,且溶剂二甲基亚砜无干扰。因此采用367 nm波长下测定喜树碱含量,并进行含量测定方法学考察。喜树碱的标准曲线方程为y=0.042 3x+0.013 2,r=0.999 6,浓度在4~20μg/mL与其吸光度呈现良好的线性关系。平均加样回收率为101.76%,RSD为0.75%,结果重现性良好,符合方法学要求。在367 nm波长下测定喜树碱含量,所受干扰较小,测定结果可信度高。The ultraviolet absorption of eamptotheein was increased at 254 nm after the laetonie ring opening,and the toxicity of camptothecin was aslo increased. In view of the current determination of camptothecin content at 254 nm,a new method content was explored for determination of camptothecin.Camptothecin had a large absorption at 367 nm from 200 nm to 400 nm, and dimethyl sulfoxide was undisturbed. Therefore,the content of camptothecin was determined at 367 nm and the methodology was investigated. The regression equation is y=0.042 3x+0.013 2,r=0.999 6. The concentration of camptothecin was in a good linear relationship with its absorbance within the range of 4~20 μg/mL. The average recovery of sample was 101.76% and RSD was 0.75%, the results had good reproducibility and more reliable.
