摘 要:目的探讨不同剂量乌司他丁(ulinastatin,UTI)对人心脏微循环内皮细胞(Human Cardiac microvascular endothelial cell,HCMEC)体外循环血清损伤的作用。方法以体外培养的HCMEC为研究细胞,分为5组:空白对照组;体外循环组;UTI大剂量组(1 000 U/m L);UTI中剂量组,(500 U/m L);(5)UTI小剂量组(100 U/m L)。每组各9个培养皿。记录各组细胞存活率;检测各组细胞上清液中NO (Nitric oxide)、ET-1(endothelin-1)的含量。结果 (1)空白对照组相对细胞存活率高于体外循环组(P<0.05),而体外循环组相对细胞存活率低于UTI各剂量组(P<0.05),UTI大剂量组相对细胞存活率明显高于UTI中小剂量组(P<0.05);(2)体外循环组NO、ET-1均高于空白对照组(P<0.05);(3) UTI各剂量组ET-1低于体外循环组(P<0.05),而NO均高于体外循环组(P<0.05);(4) UTI大剂量组NO明显高于UTI中小剂量组(P<0.05),但ET-1明显低于UTI中小剂量组(P<0.05);(5) UTI小剂量组与UTI中剂量组在NO差异无统计学意义(P>0.05)。结论(1) UTI对人体外循环血清损伤HCMEC的有一定的保护作用;(2)体外循环血清损伤HCMEC过程中,大剂量UTI较中小剂量UTI保护作用好。Objective To investigate the effects and possible mechanism of different doses of ulinastatin(UTI) on the extracorporeal circulation of human microcirculation endothelial cells in vitro. Methods After humancardiac microvascular endothelial cells(HCMEC) were cultured in vitro , experiment group was divided as follow:(1) the control group; extracorporeal circulation group; the high-dose UTI group 1000 units/mL); themiddle-dose UTI group;the small dose UTI group 100 units /ml). The percentage of cell activity in each group wasrecorded. In addition,the levels of NO、ET-1 were measured and analyzed in the cells of each group.Results(1) In the blank control group, the levels of cell survival rate was lower than the extracorporeal circulation group (P〈 0.05 ),while the relative cell survival rate in the extracorporeal circulation group was lower than that in theustendazin dose group( P〈 0.05 ) . The relative cell survival rate was significantly higher in the high-dose groupthan that in middle/small dose UTI group ( P〈 0.05) . (2) In the extracorporeal circulation group,the levels ofNO,ET-1 were all higher than the blank control group( P〈 0.05) . (3) The level of ET-1 of UTI groups werelower than that in the extracorporeal circulation group ( P〈0.05),while the level of NO of UTI groups were all higher than those in ulinastatin dose group ( P〈 0.05) . (4) In the large dose group,the level of NO wassignificantly higher than those in middle/small dose group ( P〈 0.05),but the level of ET-1 was significantly lowerthan those in middle/small dose UTI group ( P〈 0.05) . (5) There was no significant difference between themiddle dose group and the small dose group in the level of NO( P〉0.05 ) .Conclusion(1) Ulinastatin hasaprotective effect on the microvascular endothelial cells of the human heart.
