摘  要:目的:研究千根草醇提物对链脲佐菌素(STZ)致糖尿病大鼠肾损伤的保护作用。方法:向50只SD大鼠尾静脉注射链脲佐菌素(STZ)120 mg·kg^(-1),3 d后测空腹血糖(FBG)。将成糖尿病模型的大鼠随机分为:模型组、吡格列酮组(0.01 g·kg^(-1))及千根草醇提物20 g·kg^(-1)组,千根草醇提物40 g·kg^(-1)组。每组大鼠均灌胃给药,连续灌胃90 d,每天一次。另设空白组SD大鼠10只,每日灌胃给予等体积的生理盐水。正常组给予正常饲料进行喂养,其余各组均给予高脂饲料喂养。末次给药2 h后,测定大鼠的空腹血糖。采集尿液,用终点法测24 h尿白蛋白(UP)含量。全自动生化分析仪测定大鼠血清中肌酐(Scr)、尿素氮(BUN)。通过HE染色观察大鼠肾脏组织的病理学变化。试剂盒检测肾脏组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活性及丙二醛(MDA)的含量。Western blot法检测肾组织中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)的表达。结果:与模型组相比,千根草醇提物组大鼠的血糖明显降低,尿微量白蛋白含量显著降低,肾功能有明显提升(P<0.05)。肾脏组织中SOD、CAT的活性明显上升、MDA含量下降(P<0.05)。TNF-α、IL-1β、IL-6的蛋白含量显著降低(P<0.05)。结论:千根草醇提物对STZ所致糖尿病大鼠肾损害有一定的保护的作用,其机制可能为提高肾组织的抗氧化及抗炎的能力有关。OBJECTIVE To investigate the renal protective effect of ethonal extracts of Euphorbia thymifolia L.in streptozotocin(STZ)induced diabetic rats.METHODS Fifty rats were injected with STZ through tail vein(120 mg·kg-1),than the fasting blood glucose(FBG)was detected after 3 days.Diabetic rats were randomly divided into following groups:model control group,pioglitazone group(0.01 g·kg-1),ethonal extracts of Euphorbia thymifolia L.groups(20,40 g·kg-1).The rats were medicated once a day for 90 d.Ten normal rats were considered as normal control group and were given equal volume of saline.During the period,the normal rats were fed with standard rodent chow.Meanwhile,the rats of other groups were fed with high fat diet.Two hours after last medication,the FBG of rats was determined.The urine was collected for 24 h urinary albumin(UP)detection.The serum levels of creatinine(Scr)and urea nitrogen(BUN)of rats were detected using automatic biochemical analyzer.Pathological changes of kidney tissues were observed by HE staining.The activities of superoxide dismutase(SOD),catalase(CAT)and malondialdehyde(MDA)content in kidney tissues were detected.Moreover,the expressions of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)were conducted by Western blot.RESULTS Compared with model control group,the FBG and UP contents in ethanol extracts of Euphorbia thymifolia L.groups were obviously reduced,which showed that the renal function of diabetic rats was improved;the activities of SOD and CAT were significantly increased,and the MDA level was decreased(P〈0.05).In addition,the expressions of TNF-α,IL-1βand IL-6 were markedly downregulated by ethanol extracts of Euphorbia thymifolia L.(P〈0.05).CONCLUSION Ethanol extracts of Euphorbia thymifolia L.play a role in the renal protection,which is associated with improving the antioxidation and anti-inflammatory ability of renal tissues.
