摘 要:目的骨肉瘤是一类发生于骨间叶组织的恶性肿瘤,恶性程度高。当前其发病的具体分子机制尚不明确且受多种因素影响。环状RNA(circRNA)已被证实在多种疾病中发挥重要的调控作用,可以通过分子海绵机制吸附miRNA进而调控关键基因表达。本研究拟通过查找骨肉瘤circRNA表达谱芯片,并分析差异表达circRNA及其潜在调控机制,预测其在骨肉瘤发生发展过程中的作用。方法通过GEO数据库查找circRNA芯片表达谱,通过GEO2R对芯片进行差异表达分析,对筛选出的上调和下调表达最明显的circRNA各5个做内源竞争性RNA调控网络图,预测出潜在靶mRNA,并应用生物信息学分析方法分析了这些靶mRNA的功能富集情况。结果在骨肉瘤circRNA表达谱芯片(GSE96964)结果中,有110个明显差异表达的circRNA,包括8个明显上调和102个明显下调的circRNA,通过位点预测网站,在5个上调最明显的circRNA下游预测出127个mRNA,5个下调最明显circRNA下游预测出158个mRNA。OMICSbean分析结果显示,这些基因所富集的通路与骨肉瘤的发生发展有密切关系。结论本研究筛选出的110个明显差异表达circRNA不仅可以作为骨肉瘤诊断与治疗的分子标志物,而且在骨肉瘤的发生发展过程中有潜在的调控作用。Objective Osteosarcoma (OS) is a type of malignant bone tumor with a high fatality rate.However, the molecular mechanism of OS is still unclear and is influenced by many factors. Many circR- NAs have been shown to play important roles in the pathogenesis of some diseases by acting as ceRNA mechanism. In this study, we presented this research to find circRNA expression profile of OS and analyze the potential regulating function of differentially expressed circRNAs. Methods A circRNA expression profile GSE96964 were found in GEO dataset. The ceRNA analysis for 5 most up-regulated or 5 most down-regulated circRNAs were then performed. The bioinformatics methods were used to analyze the pathway enrichment of these predicted target mRNAs in order to better understand the function of circRs. NAs. Results A total of 110 differentially expressed circRNAs between OS and control were detected, including 8 up-regulated and 102 down-regulated circRNAs. The ceRNA analysis showed that 127 mR- NAs were potential target mRNAs of 5 most up-regulated circRNAs and 158 mRNAs were potential target mRNAs of 5 most down-regulated circRNAs. The pathway enrichment analysis showed that these enriched pathways involved in process of OS occurrence and development. Conclusions We found 110 differenti- ally expressed circRNAs in a circRNA expression profile of OS. These differentially expressed circRNAs could not only provide new diagnosis and treatment strategy for osteosarcoma, but also were predicted to play critical roles in process of OS occurrence and development.
