摘 要:目的:观察不同剂量虎杖苷与姜黄素对百草枯(PQ)中毒小鼠行为及炎性因子、氧化应激相关分子表达的影响。方法:将60只BALB/c小鼠随机分为正常对照组、PQ中毒组、姜黄素低剂量组、姜黄素高剂量组、虎杖苷低剂量组、虎杖苷高剂量组,每组各10只。PQ中毒组小鼠仅予20%PQ水剂以15mg/kg剂量灌胃一次性染毒,2h后予0.9%氯化钠溶液以40ml/kg剂量灌胃,1次/d,连续14d;姜黄素高、低剂量组、虎杖苷高、低剂量组染毒2h后,分别予(20mg/kg、5mg/kg)姜黄素溶液、(20mg/kg、5mg/kg)虎杖苷溶液灌胃,1次/d,连续14d;正常对照组予0.9%氯化钠溶液以75ml/kg剂量灌胃1次,2h后予0.9%氯化钠溶液以40ml/kg剂量灌胃,1次/d,连续14d。观察各组小鼠行为学特性、生存率、肺干湿比等,苏木精-伊红染色后观察肺组织切片,ELISA法测定小鼠血清中MPO、CAT、MDA、SOD水平的变化。结果:与正常对照组比较,PQ中毒组小鼠静伏少动、精神萎靡,死亡主要发生在第7~10天,肺干湿比明显增加,肺组织损伤显著,同时血清中CAT、SOD水平明显降低(P <0.01),MPO、MDA水平明显升高(P<0.01);与PQ中毒组比较,姜黄素高、低剂量组与虎杖苷高、低剂量组小鼠随给药次数增加症状明显减轻,逐渐活动、精神恢复,病死率明显下降,肺干湿比下降,肺组织损伤明显改善,血清中CAT、SOD水平升高(P<0.01),MPO、MDA水平下降(P<0.01)。此外,与同剂量姜黄素组相比,虎杖苷低剂量组MPO水平升高(P<0.01)、虎杖苷高剂量组SOD水平升高(P<0.01)。与同种药物低剂量组相比,虎杖苷高剂量组CAT、SOD水平均明显升高(P<0.01),MPO、MDA水平明显降低(P<0.01);姜黄素高剂量组CAT、SOD水平均明显升高(P<0.01),MDA水平明显降低(P<0.01)。结论:虎杖苷可通过抑制氧化应激,减轻炎性损伤,从而对PQ导致的小鼠组织损伤发挥保护作用,且虎杖苷与姜黄素的保护作用相当。虎杖苷可作为PQ中毒的潜在治疗药�Objective:To observe the effects of different doses of polydatin and curcumin on the behavior,inflammatory factors and oxidative stress-related molecules in mice with paraquat(PQ)poisoning.Method:A total of60 BALB/c mice were randomly divided into normal control group,PQ poisoning group,curcumin low-dose group,curcumin high-dose group,polydatin low-dose group,and polydatin high-dose group,10 in each group.PQ mice received only 20% PQ solution at a dose of 15 mg/kg intragastrically only once,then 2 hours later 0.9% sodium chloride solution was administered at 40 ml/kg once daily for 14 days;curcumin treatment group(high and low dose group)and polydatin treatment group(high and low dose groups)were respectively given(20 mg/kg,5 mg/kg)curcumin solution,(20 mg/kg,5 mg/kg)polydatin solution gavage after 2 hours of exposure,once a day for 14 days;The normal control group was intragastrically administered with 0.9% sodium chloride solution at 75 ml/kg,after 2 hours 0.9% sodium chloride solution was intragastrically administered at 40 ml/kg once daily for 14 days.The behavior,survival rate,and wet to dry ratio of the lungs were observed in each group.Lung sections were observed after HE staining.The levels of MPO,CAT,MDA,and SOD in the serum of mice were determined by ELISA.Result:Compared with the normal control group,the mice in PQ group were quiescent and apathetic.The death occurred mainly in 7-10 days.The wet to dry ratio was significantly increased,and the lung tissue injury was deteriorated.At the same time,serum CAT and SOD levels were significantly reduced(P〈0.01),MPO,MDA levels increased significantly(P〈0.01).Compared with PQ poisoning group,curcumin treatment group(high and low dose group)and polydatin group(high and low dose group)mice were gradually activities and mental recovery,mortality rate was reduced,and wet to dry ratio was decreased significantly and lung tissue damage improved significantly.The serum CAT,SOD levels increased(P〈0.01),MPO,MDA levels
