摘 要:为了探索眼树莲乙醇提取物和不同萃取部位的体外抗肿瘤作用,以人胃癌SGC细胞株、人肝癌BEL细胞株、慢性髓原白血病K562细胞株、宫颈癌Hela细胞株、乳腺腺癌MCF-7细胞株、肺腺癌A549细胞株为供试细胞株,用MTT法对眼树莲不同提取部位进行体外抗肿瘤活性筛选,并观察细胞形态。结果表明:乙醇提取物(A)、石油醚部位(B)、乙酸乙酯部位(C)均对SGC细胞具有明显的抑制性,IC50值分别为71.73、55.83、51.95μg/mL,而对Hela、MCF-7、A549 3种细胞有弱的抑制性。A、B部位对BEL细胞有弱抑制性,C部位对BEL细胞具有一定的抑制性,IC50值为72.62μg/mL。B、C部位对K562细胞具有明显的抑制性,IC50值分别为53.34、55.99μg/mL。正丁醇部位(D)对SGC、BEL、K562、Hela、MCF-7、A549这6种细胞均没有抗肿瘤抑制性。B、C均可使相应的肿瘤细胞株的细胞形态发生变化,引起细胞株的坏死,并呈现明显的剂量-时间-效应关系,为进一步研究眼树莲的抗肿瘤作用奠定基础。Human gastric carcinoma SGC cell line, Human hepatocellular carcinoma BEL cell line, Chronic myelogenous leukemia K562 cell line, cervical cancer Hela cell line, MCF-7 cell line of adenocarcinoma of the breast and lung adenocarcinoma A549 cell line were used to test the in-vitro anti-tumor activity of the ethanol extract, and different extract from Dischidia chinensis with different extraction methods was measured by methyl thiazolyl tetrazolium(MTT) colorimetry. The results showed that the ethanol extract(A), petroleum ether extract(B) and acetic ether extract(C) had manifest antitumor activity on SGC cell line, IC50 was 71.73, 55.83, 51.95 μg/mL, respectively. But A, B and C showed mild anti-tumor activity on Hela, MCF-7 and A549 cell line. A and B showed mild antitumor activity on BEL cell line, while C showed less antitumor activity on BEL cell line with IC50 72.62 μg/mL. B and C showed manifest antitumor activity on K562 cell line with IC50 53.34, 55.99 μg/mL, respectively. But n-butanol extract(D) did not show antitumor activity on SGC, BEL, K562, Hela, MCF-7 and A549. B and C both induced a change in the cell morphology of the corresponding tumor cell lines and tumor cell lines necrosis, and both showed dose-time-effect relationships. This research established a foundation to study further the anti-tumor effect of D. Chinensis.
