摘  要:目的 探讨紫檀芪(Pter)是否具有抗急性肾缺血再灌注损伤(IRI)作用及其可能发生机制.方法 选取雄性SD大鼠30只,随机分为假手术组(Sham组,n=10)、肾缺血再灌注组(IRI组,n=10)和紫檀芪治疗组(IRI+Pter治疗组,n=10).通过夹闭双肾动脉建立大鼠肾脏急性缺血再灌注模型,2周后检测大鼠血清肌酐、尿素氮水平,超氧化物歧化酶(SOD)活性、肿瘤坏死因子-α(TNF-α)水平以及caspase-3活性和核因子(NF-κB)的表达水平.结果 Pter能有效改善急性肾缺血再灌注损伤后肾功能,与未治疗的IRI组比较,急性肾IRI大鼠接受Pter治疗后,体内SOD活性升高(P〈0.01),TNF-α表达水平降低(P〈0.01).另外,Pter可抑制急性肾IRI诱导的caspase-3活化和NF-κB的表达(P〈0.01).结论 Pter可能通过抗氧化应激、减轻炎症反应、抗细胞凋亡和抑制TLR4/NF-κB信号传导通路起到防止急性肾IRI的作用.Objective To investigate whether Pter protects against acute renal ischemia reperfusion in rats and its possible mechanism .Methods Thirty adult male Sprague Dawley rats were randomly divided into the following 3 groups:Sham group ( n=10 ) , IRI group ( n=10 ) and IRI+Pter group ( n=10 ) .The bilateral arteries were bloked to make the IRI models .Serum nitrogen and creatinine were evaluated after 2 weeks.The concentrations of SOD and TNF-αwere measured by ELISA .The caspase 3 activity and the nuclear factor ( NF-κB ) expression were detected by immunohistochemistry . Results Pter treatment improved renal function following acute renal IRI .Compared with IRI group , SOD expression levels were significantly increased ( P 〈0.01 ) and TNF-αexpression levels were significantly decreased ( P〈0.01) following treatment with Pter in acute renal IRI rats .In addition, Pter significantly attenuated caspase-3 activity and the nuclear factor ( NF-κB ) expression ( P 〈0.01 ) . Conclusions Pter may prevent acute renal IRI through antioxidant stress , alleviating inflammation , antiapoptosis and inhibiting TLR 4/NF-κB signaling pathway .
