摘 要:以28个观赏葫芦品种为试材,采用RAPD分子标记技术,研究了其DNA多态性以及亲缘关系,以期为观赏葫芦的分子育种及资源保护和开发利用鉴定提供参考。结果表明:18个引物共扩增出111条谱带,其中102条表现出多态性,多态率为91.89%;UPGMA聚类分析表明,个体间遗传距离在0.32~0.89,在L1(GS=0.45)处,28个观赏葫芦品种可分为三大类,在L2(GS=0.722)处,可分为11类。表明葫芦品种在分子水平上呈现多态性,它们之间的亲缘关系与形态学有一定的相关性。28 cultivars of ornamental calabash were taken as materials,RAPD sequences were amplified to analysis the polymorphism and the genetic relationship of them,to provide reference for molecular breeding,resources protection and utilization identification of ornamental calabash.The results showed that total 111 bands were amplified by 18 primers,in which 102 bands were polymorphic,the percentage of polymorphic band was 91.89%.The genetic distance of individual was from 0.32 to 0.89 by UPGMA phylogenetic tree.28 ornamental calabash cultivars were divided into three subgroups at L1(GS=0.45),and they could be classified into eleven subgroups at L2(GS=0.722).It was indicated that the polymorphisms of ornamental calabash cultivars at molecular level and their phylogenetic relationships were correlated with morphology.
