摘  要:目的:研究信号调节激酶(ERK^(-P))在心力衰竭大鼠心肌细胞中含量的变化以及肉桂醛对其影响。方法:Wistar成年大鼠90只随机分为假手术组、模型组和治疗组,每组30只。将大鼠左冠状动脉的前降支结扎使其缺血造成慢性心力衰竭(CHF)模型,假手术组不结扎动脉,只把心脏挤出;治疗组造模成功后给予肉桂醛灌胃5周,利用苏木精–伊红染色法(HE)染色技术观察各组大鼠心肌的组织形态学方面变化、WB法检测各组大鼠心肌细胞中ERK^(-P)蛋白表达量的改变。结果:假手术组与模型组相比,左室峰压、左室压力最大上升和下降速率(±dp/dtmax)明显升高,心肌组织中ERK^(-P)明显降低,差异具有统计学意义(P <0.01);模型组与治疗组相比,左室峰压、左室压力最大上升和下降速率(±dp/dtmax)明显降低,心肌中ERK^(-P)比模型组明显下降,差异具有统计学意义(P <0.01),相比假手术组ERK^(-P)升高,差异具有统计学意义(P <0.05)。结论:在CHF发生过程中ERK^(-P)起着非常重要的作用,肉桂醛具有减少CHF大鼠心肌细胞中ERK^(-P)含量的作用,所以能够保护和改善心力衰竭大鼠心功能。Objective To study the changes in the content of signal regulated kinase(ERK-P) in cardiac myocytes of rats with heart failure and the effect of cinnamaldehyde on it. Methods 90 adult Wistar rats were randomly divided into the sham operation group, the model group and the treatment group, with 30 rats in each group. Ligation of the left anterior descending branch of the left coronary artery made the model of chronic heart failure induced by ischemia.The sham operation group did not ligate the artery, only the heart was extruded. The treatment group was given cinnamaldehyde after successful modeling. After 5 weeks of gavage, morphological changes of myocardium in rats were observed by HE staining, and the expression of ERK-P protein in rat cardiac myocytes was detected by WB. The results of the sham operation group and model group, left ventricular pressure maximal rate of increase and decrease(+dp/dtmax) and firmly left pressure, significantly increased ERK-P in myocardium significantly decreased(P〈0.01); compared with the model group and treatment group, left ventricular pressure, firmly pressure rising and falling rate(dp/dtmax) significantly reduced myocardial ERK-P significantly decreased than the model group(P〈0.01), compared with the sham operation group(P〈0.05) ERK-P. Conclusion ERK-P plays a very important role in the development of chronic heart failure. Cinnamaldehyde can reduce the content of ERK-P in myocardial cells of chronic heart failure rats, so it can protect and improve heart function in rats with heart failure.
