摘 要:目的观察白藜芦醇、染料木黄酮、姜黄素和大黄素对细胞衰老及细胞生长增殖能力的影响。方法配置白藜芦醇、染料木黄酮、姜黄素和大黄素不同浓度的DMEM培养基,分别培养人胚肺二倍体成纤维细胞(2BS),观察2BS细胞形态、体外传代次数,以MTT法观察细胞生长增殖能力变化,以单细胞凝胶电泳(彗星试验)法观察细胞DNA单链断裂水平。结果低浓度白藜芦醇、染料木黄酮、姜黄素和大黄素DMEM培养基培养的细胞体外传代次数均较对照组细胞下降;上述抗氧化剂在低浓度时可促进细胞生长增殖,但在高浓度时抑制细胞生长增殖;四种抗氧化剂均可断裂单链DNA,且随浓度加大损伤作用愈大。结论白藜芦醇、染料木黄酮、姜黄素和大黄素具有促进细胞生长增殖和细胞毒性双重作用,其细胞毒性源于其对DNA损伤作用。Objective To investigate the effects of resveratrol,genistein,curcumin and allicin on the replicative senescence growth and proliferation. Methods The morphology,population doublings(PDs) in vitro,growth and proliferation,and the single-strand DNA breakage levels of human embryonic lung diploid fibroblasts(2 BS) cultured with DEME supplemented with resveratrol,genistein,curcumin or allicin were observed.Results The PDs of the cells incubated with resveratrol,genistein,curcumin or allicin in low concentration were less than those of control cells. The antioxidants above in low dose promoted the growth and proliferation of the cells,whereas,they inhibited the growth and proliferation of 2BS in high dose.The antioxidants above broke DNA strand,moreover,the breakage of DNA strand increased with the antioxidant concentration. Conclusions Resveratrol,genistein,curcumin and allicin have the double actions to promote proliferation and DNA strand breakage. The cytotoxicity of the antioxidants results from the damage effect of DNA.
