摘 要:为了筛选绵羊EPAS1基因SNP多态位点,本文设计2对引物利用PCR技术从5个绵羊品种(3个高原品种和2个低地品种)基因组中扩增了EPAS1基因exon12和exon16片段,测序结果发现高原绵羊和低地绵羊群体间存在4个SNPs位点,其中1个位于第12外显子(276 T>C),3个位于第16外显子(904 T>C、914A>C和977IndelA),这些多态性位点可能对EPAS1基因的功能具有重要的调控作用,这为藏绵羊高原低氧适应机制研究提供了基础。In order to screen SNP polymorphic loci of the sheep EPAS1 gene, two pairs of primers were designed to amplify Exon12 and Exon16 fragments of EPAS1 gene from genomes of five sheep varieties(three highland varieties and two lowland varieties) using PCR technology. The sequencing results showed there were four SNPs between the Tibetan and lowland sheep, in which, one SNP(276 TC) was in exon 12, and three SNPs(904 TC, 914 AC and 977 IndelA) in exon 16. These polymorphisms may have important regulatory effects on the function of EPAS1 gene, which provided a basis for study of the adaptation mechanism of hypoxia in sheep.
