摘 要:为明确本课题组收集的香稻资源的香味基因突变类型,及其杂种F3代的多基因聚合情况,本研究开展了香味表型鉴定和9个基因的基因型鉴定,鉴定基因包括香味基因Badh2、抗稻瘟病基因Pita和Pib、抗白叶枯病基因Xa21和Xa23、抗褐飞虱基因Bph14和Bph15、耐涝基因Sub1、抽穗期耐冷基因Ctb1。结果表明:本课题组收集的香稻材料中12份属于Badh2第7外显子8个碱基缺失和3个碱基突变的类型。传统感官评判方法和分子标记方法鉴定香稻亲本的结果能互相印证。40个杂种F3代单株中,16F1051-1株系的1-4、1-5、1-8和1-10这4个单株成功聚合了Badh2、Xa21、Pita、Pib、Bph14、Bph15和Sub1共7个基因;共计筛选到聚合至少5个基因的单株17个,为后续优质香稻育种提供了可供借鉴的经验和多抗育种新材料。In order to clarify the aroma gene mutation type of aromatic rice collected by our research group and the gene pyramiding in F3 generation of aromatic rice, we carried out the phenotyping of aroma and genotyping of nine important genes, including aroma gene Badh2, blast resistance gene Pita and P/b, bacterial blight resistance gene Xa21 and Xa23, brown planthopper resistance gene Bphl4 and Bphl5, submergence tolerance gene Subl, and heading stage cold tolerance gene Ctb 1. The results showed that twelve accessions of aromatic rice collected by our research group belonged to the 8 bp deletion and 3 bp mutation type in exon 7 ofBadh2 gene. The results of traditional sensory evaluation and molecular marker identification could be verified by each other. Among the 40 individuals of F3 generation from aromatic flee, four individuals, marked as plant 1-4, 1-5, 1-8 and 1-10 of line 16F1051-1, were successfully pyramided seven genes including Badh2, Xa21, Pita, Pib, Bph14, Bph15 and Sub 1. A total of seventeen individuals which could at least pyramid five genes were screened out. This study provided successfully experiences and multi-resistant aromatic lines for the future aromatic rice breeding.
