摘  要:筛选羊肉、猪肉及鸭肉基因组DNA提取方法,并进行DNA条形码鉴定.以羊猪鸭三种肉类为实验材料,采用传统法(SDS法)、chelex-100法、异硫氰酸胍法、试剂盒法提取基因组DNA,并用琼脂糖凝胶电泳、紫外分光光度法和COI序列扩增法对提取的效果进行检测和比较,利用建立的方法提取DNA并进行DNA条形码鉴定.四种方法均能从样品肌肉组织提取到基因组DNA,其中SDS法和试剂盒法得到的DNA质量最佳;异硫氰酸胍法DNA质量较好,操作时间长; chelex-100法简单、快速,但DNA含杂质较多,质量较差.四种方法均可提供满足DNA条形码分子鉴定的要求DNA,实际应用中可根据条件选择相应的方法.To establish a universal DNA extraction method for future research on authentica- tion of mutton, pork, and duck by DNA barcoding. Genomic DNA of musclar tissues from mutton, pork,and duck were extracted through SDS method , GuSCN method, helex- 100 method and Kit respectively. The DNA was detected by optical density, agarosegel electropho- resis and COI sequence amplification. The optimized method was used to extract DNA from animal musclar tissue for species identification. The SDS method , GuSCN method, chelex - 100 method and Kit could produce genomic DNA, among them, SDS method and Kit were best. The established extraction methods were proved to be suitable for DNA barcoding re- search. Suitable method can be choose to genomic DNA of muscle tissue according to the ac- tual conditions of the laboratory.
