摘 要:目的探讨胸腺素β4对深Ⅱ°烫伤创面愈合的影响及作用机制。方法选取健康SD大鼠40只,将大鼠随机分为对照组、低剂量组、中剂量组和高剂量组,每组10只,其中对照组、低剂量组、中剂量组和高剂量组分别给予生理盐水、2、6和18μg胸腺素β4处理创面,观察各组创面愈合情况,采用ELISA法检测组织IL-10和TNF-α水平,采用Western blot检测创面VEGF、Caspase-3、NF-κB表达,采用RT-PCR检测创面VEGF mRNA、TGF-β1 mRNA表达。结果高剂量组烫伤后7、14 d大鼠创面愈合率分别为(40.06±1.67)%和(70.03±1.88)%,明显高于对照组、低剂量组和中剂量组(P<0.05);高剂量组烫伤后12、24和48 h时IL-10均明显高于对照组、低剂量组和中剂量组(P<0.05),而TNF-α明显低于对照组、低剂量组和中剂量组(P<0.05);高剂量组烫伤后7、14 d时VEGF明显高于对照组、低剂量组和中剂量组(P<0.05),而Caspase-3和NF-κB明显低于对照组、低剂量组和中剂量组(P<0.05);高剂量组烫伤后7、14 d时VEGF mRNA和TGF-β1 mRNA表达明显高于对照组、低剂量组和中剂量组(P<0.05)。结论胸腺素β4可促进大鼠深Ⅱ°烫伤创面愈合,其机制可能与降低炎性因子、Caspase-3及NF-κB水平,提高VEGF表达有关。Objective To investigate the effect and mechanism of thymosin β4 on healing of deep second degree scaldwounds. Methods Forty healthy SD rats were randomly divided into control group, low dose group, middle dose group and high-dose group, with 10 rats in each group. Each of these groups was treated with normal saline, 2 μg, 6 μg and 18 μg thymosin β4respectively. The wound healing of each group was observed. The levels of IL-10 and TNF-α were detected by ELISA. Westernblot was used to detect the expressions of VEGF, Caspase-3 and NF-κB, and RT-PCR was used to detect the expressions ofVEGF mRNA and TGF-β1. Results The wound healing rates at 7 d and 14 d after scald in high dose group were (40. 06依1. 67)%and (70. 03依1. 88)% respectivley, which were significantly higher than those in the other three groups (P〈0. 05). The level ofIL-10 at 12 h, 24 h and 48 h after scald in high dose group was significantly higher than that of the other three groups (P〈0. 05),but TNF-α was significantly lower (P〈0. 05). VEGF at 7 d and 14 d after scald in high dose group was significantly higher thanthat of the other three groups (P〈0. 05), while Caspase-3 and NF-κB were significantly lower (P〈0. 05). In high dose group,the expressions of VEGF mRNA and TGF-β1 mRNA at 7 d and 14 d after scald were significantly higher than those in controlgroup, low dose group and middle dose group (P〈0. 05). Conclusion Thymosin β4 can promote the healing of deep second degreeburn wounds in rats, and the mechanism may be related to the regulation of VEGF, Caspase-3 and NF- κB.
