京尼平苷对INS-1细胞增殖与凋亡的影响

摘　　要：目的观察京尼平苷对大鼠胰岛β细胞系(INS-1)增殖及凋亡的影响。方法用CCK-8法测定细胞增殖,用毒胡萝卜素处理细胞后,用流式细胞仪测定细胞凋亡率,并用CCK-8法测定细胞活力。结果京尼平苷同胰高血糖素样肽1(GLP-1)一样能促进INS-1的增殖,并有浓度依赖性;京尼平苷同GLP-1一样能抑制毒胡萝卜素处理后的细胞凋亡,并有浓度依赖性;同时京尼平苷能抑制毒胡萝卜素对细胞的毒性作用。结论京尼平苷能促进INS-1细胞增殖,抑制INS-1细胞凋亡。Objective To study the effect of geniposide on proliferation and apoptosis of islet cell line （INS-1） in rat islet.Methods The cell proliferation was determined by CCK- 8 method,and the cell apoptosis rate was determined by the flow cytometry,and thecell viability was determined by CCK-8 method.Results Geniposide （1μmol/L-100μmol/L）,as same as glucagon like peptide 1（GLP-1）,could promote the proliferation of INS -1 cells and showed a concentration dependence.Thapsigargin may induce INS 1cells apoptosis.Treatment pancreatic INS -1 cells with geniposide or GLP- 1 could attenuate thapsigargin induced beta cell apop -tosis.Geniposide （10μmol/L） could inhibit the toxic effect of thapsigargin on cells.Conclusion Geniposide can promote proliferation ofINS- 1 cells and inhibit apoptosis of INS -1 cells.
