摘　　要：目的:通过检测瞬时受体电位离子通道A1(Transient receptor potential channel Al,TRPAl)蛋白及mRNA在冠心病患者外周血单核细胞中的表达,分析TRPA1mRNA转录水平与Gensini积分的相关性。方法:根据冠状动脉(冠脉)造影结果将入院患者分为冠心病组(39例)和非冠心病组(36例),分离提取鉴定患者外周血单核细胞,通过Western blotting、RT-PCR分别检测两组患者单核细胞TRPA1的蛋白及mRNA表达水平。根据患者是否合并高血压、糖尿病,将冠心病患者分为高血压组与非高血压组、糖尿病组与非糖尿病组,分别比较两组间TRPA1mRNA转录水平。分析冠心病患者单核细胞TRPA1mRNA转录水平与冠脉病变Gensini积分的相关性。结果:(1)成功分离提取患者外周血单核细胞,经流式细胞术鉴定,单核细胞纯度在86.1%。(2)Western Blot显示,冠心病组患者单核细胞TRPA1蛋白表达水平高于非冠心病组(1.524±0.426∶0.590±0.275,P<0.01)。(3)RT-PCR显示,冠心病组TRPA1mRNA转录水平高于非冠心病组(9.121±1.828∶3.923±2.535,P<0.01),高血压组TRPA1mRNA转录水平高于非高血压组(9.812±1.180∶8.740±1.586,P<0.05),糖尿病组TRPA1mRNA转录水平高于非糖尿病组(9.685±0.932∶8.982±0.756,P<0.05)。(4)冠心病患者TRPA1mRNA转录水平与Gensini积分呈正相关(r=0.964,P<0.01)。结论:冠心病患者单核细胞TRPA1表达水平增高,TRPA1mRNA转录水平可作为评价冠脉病变程度的新指标。Objective：To analyze the correlation between Transient receptor potential channel Al（TRPAl）mRNA transcription level and Gensini score by detecting the expression of TRPAl protein and mRNA in peripheral blood monocytes in patients with coronary heart disease（CHD）.Method：According to the results of coronary angiography,the admission patients were divided into CHD group（n=39）and non-CHD group（n=36）.Peripheral blood monocytes were isolated and identified,and the expression levels of TRPA1 in monocytes were detected by Western blotting and RT-PCR respectively.According to whether combined with hypertension and diabetes,the CHD patients were divided into hypertension group and non-hypertension group,diabetes group and non-diabetic group,and the TRPA1 mRNA transcriptional levels between the two groups was compared respectively.The correlation between the transcription of TRPA1 mRNA in monocytes and the Gensini score in coronary lesions was analyzed.Result：（1） Peripheral blood monocytes were successfully isolated and extracted.The purity of monocytes was 86.1% after flow cytometry.（2） Western Bloting showed that the expression of TRPA1 protein in monocytes in CHD group was higher than that in non-CHD group（1.524±0.426 vs.0.590±0.275,P〈0.01）.（3） RT-PCR showed that the transcription level of TRPA1 mRNA in CHD group was higher than that in non-CHD group（9.121±1.828 vs.3.923±2.535,P〈0.01）,and TRPA1 mRNA in hypertension group was higher than that in non-hypertension group（9.812±1.180 vs 8.740± 1.586,P〈0.05）,the transcription level of TRPA1 mRNA in diabetic group was higher than that in non-diabetic group（9.685±0.932 vs.8.982±0.756,P〈0.05）.（4） There was a positive correlation between TRPA1 mRNA transcription level and Gensini score in patients with CHD（r=0.964,P〈0.01）.Conclusion：The expression level of TRPA1 in monocytes is increased in patients with CHD,and the level of TRPA1 mRNA transcription can be used as a new indicator to evaluate the severity
