摘 要:目的探讨治疗用卡介苗(简称BCGt)对MB49膀胱肿瘤细胞(简称MB49细胞)增殖的影响。方法体外培养MB49细胞,每天观察并采用CCK-8法检测细胞生长情况,绘制细胞生长曲线。取对数生长期的MB49细胞与不同浓度(20、15、10、8、6、3、2. 5、2、1. 5、1 mg/mL)的BCGt作用72 h,CCK-8法检测细胞增殖能力,计算细胞抑制率。同时采用ELISA法测定细胞培养液中IL-6、IL-8及IL-15的浓度。结果 MB49细胞于第1~4天处于对数生长期,细胞增殖速度较快,细胞形态正常。BCGt可有效抑制MB49细胞的增殖,且呈剂量依赖性,IC_(50)为3. 650 mg/mL。经BCGt作用后细胞培养液中IL-6和IL-15浓度明显增高(P <0. 05),IL-8浓度未发生明显变化(P> 0. 05)。结论BCGt可抑制MB49细胞的增殖,可能是通过提高IL-6和IL-15的水平,促进细胞免疫应答,从而发挥抗肿瘤作用的。Objective To evaluate the effect of BCG for therapeutic use(BCGt)on proliferation of bladder tumor MB49 cells. Methods MB49 cells were cultured in vitro and determined for growth by CCK-8 method daily,based on which a cell growth curve was plotted. The MB49 cells in logarithmic growth phase were treated with BCGt at various concentrations(20,15,10,8,6,3,2. 5,2,1. 5 and 1 mg/mL)for 72 h and determined by CCK-8 method,based on which the inhibitory rate of cell growth was calculated. Meanwhile,the concentrations opf IL-6,IL-8 and IL-15 in cell culture medium were determined by ELISA. Results The MB49 cells were in logarithmic growth phase within 1 ~ 4 d after culture,which grew rapidly in normal shape. BCGt inhibited the proliferation of MB49 cells effectively in a dosedependent manner,of which the IC_(50) was 3. 650 mg/mL. The concentrations of IL-6 and IL-15 in medium of cells after treatment with BCGt increased significantly(P〈0. 05),while that of IL-8 showed no significant change(P〉0. 05).Conclusion BCGt inhibited the proliferation of MB49 cells by a possible mechanism of increasing the IL-6 and IL-15 levels and promoting cellular immune response.
