摘 要:目的观察破壁与非破壁处理灵芝孢子粉的化学成分释放及免疫调节作用的差异。方法应用高效液相色谱-质谱(HPLC-MS)测定两种方法处理的灵芝孢子粉中灵芝酸A、C2、D的含量,采用环磷酰胺制备免疫功能低下动物模型,通过检测小鼠存活率和体重、免疫器官指数、股骨有核细胞数、淋巴细胞增殖转化率、免疫球蛋白水平,及外周血中CD3+、CD4+和CD8+数量,全面评价两种方式处理的灵芝孢子粉的免疫调节作用。结果经破壁处理的灵芝孢子粉能提高小鼠存活率,显著升高股骨有核细胞数,且能抑制环磷酰胺导致的T淋巴细胞及B淋巴细胞增殖转化率的降低,提高小鼠血清中免疫球蛋白水平,增加免疫低下小鼠的淋巴细胞数量。结论经破壁处理的灵芝孢子粉对环磷酰胺免疫低下小鼠的免疫功能有较明显的促进作用。Objective To observe the differences of the release of chemical components and immunomodulatory effects between the wall-broken and wall-unbroken Ganoderma spore powder. Methods HPLC-MS was developed to identify and quantify the contents of ganoderic acid A, C2 and D in Ganoderma spore powder. Cyclophosphamide-induced immunosuppressive mice model was applied to evaluate the immunomodulatory effects. Body weight, survival rate, immune organ index, femur nucleated cell number, lymphocyte proliferation and transformation rate, immunoglobulin levels, and the number of CD3+, CD4+ and CD8+ in peripheral blood were measured and analyzed. Results The wall-broken Ganoderma spore powder improved the survival rate, increased the femur nuclear cell number and the thymus index compared to the model. Furthermore, the wall-broken Ganoderma spore powder extenuated the suppressed proliferation and transformation rate of T and B lymphocytes, increased the serum levels of immunoglobulin and the number of lymphocytes in the immunocompromised mice. Conclusion The wall-broken Ganoderma spore powder has obvious promoting effect on the immune function of immunocompromised mice.
