摘 要:目的:建立沙菊茶水溶性成分的高效液相色谱(HPLC)指纹图谱,并测定绿原酸、表儿茶素、葛根素、木犀草苷、木犀草素、西红花苷Ⅰ和西红花苷Ⅱ等7个成分的含量及不同泡茶时间的溶出度。方法:采用HPLC法,色谱条件:色谱柱为Intersil C_(18)柱(250mm×4. 6mm,5μm);流动相为乙腈-0.085%磷酸水溶液,使用梯度洗脱法;流速为1. 0m L/min;检测波长为270 nm和440 nm;柱温为40℃。结果:1)通过中药色谱指纹图谱相似度评价系统(2004A)确定沙菊茶水溶性成分的HPLC指纹图谱共有模式,标定40个共有峰,相似度为0. 952~0. 993;2) 10批制剂中绿原酸、表儿茶素、葛根素、木犀草苷、木犀草素、西红花苷Ⅰ和西红花苷Ⅱ的含量(mg/g)分别为0. 1717、0. 3726、0. 8735、0. 0282、0. 5314、0. 5652和0. 1275 mg/g,RSD分别为1. 44%、0. 83%、0. 760%、1. 07%、0. 81%、0. 91%和1. 88%;3)茶中7个成分的含量随浸泡时间的延长逐渐升高,在60分钟后均能达到峰值。结论:HPLC指纹图谱方法与7个成分的含量检测方法以及溶出度测定方法简便、稳定、科学,可用于沙菊茶的质量控制并指导患者正确使用。Objective: To establish a HPLC fingerprint of aqueous components of Shaju tea, and detect thecontents of seven kinds of ingredients including chlorogenic acid, epicatechin, puerarin, galuteolin, luteolin, crocinⅠand crocinⅡ in the preparations and the dissolution rates of these ingredients in different soaking time. Methods:The high performance liquid chromatography (HPLC) analysis was performed on such chromatographic conditions:chromatographic column was IntersilC18 (250 mm×4.6 mm,5 μm), the mobile phase was a mixture of acetonitrileand 0.085 % phosphoric acid solution in gradient elution; the flow rate was 1.0 mL/min, and the detectionwavelength was 270 nm and 440 nm, the column temperature was set at 40℃ . Results:1) Under the abovechromatographic conditions, HPLC fingerprint common modes of aqueous components of Shaju tea were confirmedby chromatographic fingerprint similarity evaluation system(2004A), and 40 chromatographic peaks were composed,and the overall similarity degree was from 0.952 to 0.993; 2) The average contents of chlorogenic acid, epicatechin,puerarin, galuteolin, luteolin, crocinⅠand crocinⅡ were 0.1717, 0.3726, 0.8735, 0.0282, 0.5314, 0.565 2 and 0.1275 mg/g in ten batches of Shaju tea, RSDs were 1.44%, 0.83%, 0.760%, 1.07%, 0.81%, 0.91% and 1.88%; 3) Thecontents of seven kinds of ingredients raised gradually as the soaking time prolonged, and it will reach the peak valuein 60 minutes. Conclusion: The method of HPLC fingerprint, the method of detecting the contents of seven kinds ofingredients and the detection method of dissolution rate are simple, stable and scientific, and they could be used forquality control of Shaju tea and to guide the patients' right use.
