摘  要:目的:探讨除湿通痹方对大鼠急性痛风性关节炎的抗炎作用机制。方法:将72只SD大鼠随机分为空白组,模型组,阳性组,除湿通痹方高、中、低剂量组共6组,每组12只。采用踝关节腔内注射尿酸钠的方法建立大鼠急性痛风性关节炎模型,连续灌胃7 d后观察造模后各组大鼠关节肿胀度,并检测关节软组织内炎症细胞因子白细胞介素-1β(IL-1β)、IL-1R的表达。结果:各给药组炎症细胞计数、IL-lβ、IL-1R表达水平显著低于模型对照组(均P <0. 05或P <0. 01),其中高剂量组各炎症指标表达水平显著低于阳性组(均P <0. 05)。结论:除湿通痹方能改善动物模型急性痛风性关节炎的炎症反应。其作用机制可能是通过阻断关节软组织内IL-1β和IL-lR的表达。Objective To explore the anti-inflammatory mechanism of Chushitongbi prescription on rats with acute gouty arthritis. Methods Seventy-two SD (72) rats were divided into 6 groups randomly:blank group, colchicine group,model group, Chushitongbi high-dose group, middle-dose group and low-dose group, and 12 rats were included in each group. Acute gouty arthritis model of was replicated by intra-articular injecting monosodium urate crystal suspension inside the ankle joint. Rats were gavaged once a day for 7 days and joint swelling were observed. IL-l β and IL-1R expression levels in soft tissues of joint were detected. Results Inflammatory cell counts and expression level of IL-l β and IL-1R in each group were significantly lower than those in the model group (P<0.05 or P 〈0.01), the inflammation index of expression of Chushitongbi high-dose group was significantly lower than that of colchicine group (P<0.05). Conclusion The inflammatory effects induced by acute gouty arthritis in rats were able to be improved by treating with Chushitongbi prescription and the mechanism of which may be associated with the down-regulation of IL-l β and IL-1R in joint soft tissues.
