摘 要:目的探讨内脂素对大鼠肾小球系膜细胞(GMCs)细胞外基质表达的影响。方法体外培养GMCs,随机分为对照组和不同浓度内脂素(25、50、100、200 ng/mL)干预组,逆转录PCR(RT-PCR)检测GMCs纤维连接蛋白(FN)和Ⅰ型胶原(COLⅠ)mRNA,Western印记测定FN、COLⅠ蛋白。结果不同浓度内脂素干预GMCs 24 h后,细胞FN及COLⅠ的mRNA和蛋白表达均较对照组增多(P<0.05),且随着内脂素浓度增加,其表达呈逐渐增加的趋势(P<0.05)。结论内脂素呈一定的浓度依赖性促进GMCs细胞外基质表达,可能参与糖尿病肾病的发生发展。Objective To investigate the effect of visfatin on the expression of extracellular matrix in rat mesangial cells (GMCs). Methods GMCs were cultured in vitro and randomly divided into control group and intervention group with different concentrations of visfatin(25 ng/mL,50 ng/mL,100 ng/mL, 200 ng/mL). Reverse transcription-PCR(RT-PCR) was used to detect GMCs fibronectin(FN) and type Ⅰ collagen,(COLI) mRNA,Western blotting to determine FN,COLI protein. Results After treatment with different concentrations of lipoprotein for 24 hours, the mRNA and protein expressions of FN and COLI increased compared with the control group(P〈0.05),and the expression increased gradually with the increase of visfatin concentration(P〈0.05). Conclusion The visfatin promotes the expression of extracellular matrix of GMCs in a concentration-dependent manner, which may be involved in the development of diabetic nephropathy.
