摘 要:目的:探讨热量控制8周对于胰岛素抵抗肥胖大鼠糖脂代谢及下丘脑POMC蛋白表达量的影响。方法:选用SPF级8周龄大鼠50只,随机分为两组:空白组20只,普通饲料喂养;肥胖组30只,高脂饲料喂养。8周喂养后行造模检测,将肥胖组中符合造模条件的大鼠随机分为模型组、热量控制组(热控组),每组各10只,空白组中随机挑选10只组成正常组。模型组行高脂饲料喂养,热控组每日高脂饲料喂养量为模型组的70%,正常组仍按普通饲料喂养,干预时间均为8周。实验结束后处死大鼠,检测各组大鼠体质量、总胆固醇(TC)、甘油三酯(TG)、空腹血糖(FBG)、餐后血糖(PBG)和胰岛素(INS)水平变化。用Western Blotting法检测下丘脑POMC蛋白表达水平。结果:8周干预后,肥胖组大鼠GIR60~120明显低于空白组大鼠(P〈0.01)。与正常组比较,模型组大鼠的体质量明显增加(P〈0.05),并且TC、TG、PBG和INS的含量均显著升高(P〈0.01),但FBG的含量差异无统计学意义(P〉0.05),下丘脑POMC蛋白含量减少(P〈0.05)。与模型组比较,热控组大鼠的体质量显著降低(P〈0.01),TG含量有所下降(P〈0.05),且TC、PBG、INS含量均显著下降(P〈0.01),FBG的含量差异无统计学意义(P〉0.05),大鼠下丘脑POMC蛋白含量显著增多(P〈0.01)。结论:热量控制8周可以有效地控制胰岛素抵抗肥胖大鼠的体质量增长,并且改善胰岛素抵抗大鼠的糖脂代谢状况,其机制可能与下丘脑POMC蛋白的表达增多有关。Objective:To observe the effects of calorie restriction 8-week on the level of glycolipid metabolism and hypothalamic POMC protein expression in obesity and insulin resistance rats. Methods:Fifty SPF grade 8-week-old rats were randomly divided into two groups:20 in the blank group and 30 in fat group. Rats in blank group were fed with standard diet,high fat diet would be used in rats in fat group. After the intervention,obesity and IR rats were randomly divided into model group and calorie restriction group,10 in each group,the blank group randomly selected 10 only to form the control group. The control group continued to feed the standard diet while high-fat diet were kept on feeding model group. Diet supplied for the calorie restriction group was 70% of the model group. The intervention time was 8 weeks. After the experiment,the rats were sacrificed and serum level of the body weight,total cholesterol(TC),triglyceride(TG),fasting blood glucose(FBG),postprandial blood glucose(PBG)and insulin(INS)were measured. Expression level of POMC protein in hypothalamus was determined by Western Blotting. Results:After 8 weeks of intervention,GIR 60-120 in obese rats was significantly lower than that in blank rats(P〈0.01). Compared with the control group,the body weight of the model group increased significantly(P〈0.05),and the levels of TC,TG,PBG and INS were significantly increased(P〈0.01),at the same time,there was no difference in the level of FBG(P〉0.05). Expression level of POMC in the hypothalamus decreased(P〈0.05). Compared with model group,the body weight of rats in thermal control group decreased significantly(P〈0.01),the TG level of calorie restriction group decreased(P〈0.05),while the level of TC,PBG and INS decreased significantly(P 〉0.05).
