清脂通脉颗粒最佳药物剂量配比筛选与疗效验证实验研究

摘　　要：目的：探讨清脂通脉颗粒的最佳药物剂量配比,研究其治疗动脉粥样硬化对的机制与NPC1、NPC1L1之间的联系。方法：通过一次性腹腔注射维生素D3（60万IU/kg）,复合高脂饲料喂养8周来建立高脂血症大鼠模型,以构成清脂通脉颗粒的5味中药为研究对象,采用均匀设计＂5因子10水平＂的分组设计。清脂通脉颗粒灌胃4周后,以血脂四项的水平为筛选指标,筛选出最佳剂量配比组。之后将最佳配比组与空白组、模型组对照,检测肝脏与小肠中NPC1、NPC1L1的mRNA及蛋白表达。结果：（1）血脂四项检测结果显示与模型组相比,组8与组12四项结果均有统计学意义,与其他中药组相比,降脂效果更佳,故选取组8与组12的药物配比为最佳剂量配比。（2）实验结果显示,清脂通脉颗粒可以显著降低大鼠血脂水平,升高NPC1的mRNA和蛋白表达;降低NPC1L1的mRNA和蛋白表达。结论：运用均匀设计筛选获得的8组与12组的计量比例为最佳配比;清脂通脉颗粒可以通过升高NPC1的mRNA和蛋白表达,降低NPC1L1的mRNA和蛋白表达来防治动脉粥样硬化。Objective：To investigate the optimal dosage of Qingzhi Tongmai Granules and to study the relationship between the mechanism of treating atherosclerosis and NPC1 and NPC1 L1. Methods：Hyperlipidemia rat models were established by intraperitoneal injection of vitamin D3（600,000 IU/kg）and fed with high-fat diet for 8 weeks. The study consisted of five herbs that constituted QingzhiTongmai Granules,design a＂5 factor 10 level＂grouping design. After Qingzhi Tongmai Granules was intragastrically administrated for 4 weeks,the level of four items of blood lipid was used as the screening index to screen out the best dose matching group. After that,the best matching group was compared with the blank group and the model group,and the mRNA and protein expression of NPC1 and NPC1 L1 in liver and small intestine were detected. Results：（1）The results of four items of blood lipid tests showed that compared with the model group,the results of group 8 and group 12 were statistically significant. Compared with other traditional Chinese medicine groups,the lipid-lowering effect was better. Therefore,the drugs of group 8 and group 12 were selected. The ratio was the best dose ratio.（2） The experimental results showed that Qingzhi Tongmai Granules can significantly reduce the blood lipid levels in rats,increase the mRNA and protein expression of NPC1,and decrease the mRNA and protein expression of NPC1 L1. Conclusion：The proportion of 8 groups and 12 groups obtained by uniform design screening is the best ratio;Qingzhi Tongmai Granules can prevent and treat atherosclerosis by increasing mRNA and protein expression of NPC1;decreasing mRNA and protein expression of NPC1 L1.
