摘 要:目的:建立不同品种及商品地水蛭、地龙中腐胺的含量测定方法,为水蛭、地龙的质量评价提供科学的依据。方法:采用反相高效液相色谱法,Diamonsil C18(200 mm×4.6mm,5μm)色谱柱,以甲醇-水(58∶42)为流动相,流速为1.0 mL·min^-1,检测波长230 nm,柱温为室温,以腐胺为特征变量,采用SPSS19.0统计软件进行方差分析和聚类分析。结果:不同品种、规格的地龙、水蛭商品药材中腐胺成分含量不同,沪地龙和广地龙腐胺含量平均值分别为0.0406 mg·g^-1、0.0161 mg·g^-1,有显著性差异(*P〈0.05),蚂蟥含量平均值为0.1008 mg·g^-1,水蛭的含量平均值0.0681 mg·g^-1,无显著差异;聚类分析结果显示广地龙百分百聚为一类,水蛭和蚂蟥有交叉聚类现象。结论:高效液相色谱法测定地龙、水蛭中腐胺的含量,方法简便,结果准确,结合方差分析和聚类分析,为地龙、水蛭的质量评价和资源利用提供理论基础。Objective:To determinate the contents of putrescine in the different species and specifications of Pheretima and Hirudo,and provide scientific evidence for quality evalution of the medicine. Methods:RP-HPLC method was used. The column was Diamonsil C18(200 mm×4.6 mm,5 μm);Mobile phase was methanol-water(58 ∶ 42),the flow rate was 1.0 mL/min,detection wavelength was 230 nm and column temperature was room temperature. Variance analysis and cluster analysis was applied to study characteristic of Pheretima and Hirudor based on the data of analysis of the content of putrescine. Results:There are certain differences in Pheretima and Hirudo. The average putrescine content of different varieties Pheretima were 0.0406 mg·g^-1,0.0161 mg·g^-1 respectively. Significant difference was found(P〈0.05). The contents in different varieties Hirudo were 0.1008 mg·g^-1 and 0.0681 mg·g^-1. There was no significant difference between different varieties Hirudo. The result of cluster analysis showed that the percentile of Guangdilong is one kind. There are the phenomenon of cross-clustering between Hirudo. Conclusion:High performance liquid chromatography(HPLC)was used to determine putrescine in the different species and specifications of Pheretima and Hirudo,which was simple and accurate. Combining with variance analysis and clustering analysis,which provide theoretical basis for quality evaluation and resource utilization ofPheretima and Hirudo.
