摘 要:目的:探讨神经元调控因子NMDAR与GABAR在升降散治疗大鼠神经病理性疼痛中的作用,并进一步研究是否会因升降散治疗影响记忆力。方法:构建大鼠右侧改良坐骨神经慢性缩窄性损伤(i CCI)经典动物模型,采用不同剂量的升降散给i CCI模型大鼠进行灌胃处理,在给药21天,对脊髓组织中NMDAR和p-NMDAR蛋白含量,及GABAR mRNA表达水平进行检测,并在给药不同时间点定时采用水迷宫实验测试大鼠穿越平台次数。结果:与正常组相比,模型组大鼠p-NMDAR水平明显P 〈0. 05升高,GABAR mRNA表达水平明显下降;经给药处理,高剂量组抑制了pNMDAR水平升高和GABARmRNA表达水平下降,且与模型组存在显著P 〈0. 05差异,但对穿台次数无明显影响。结论:升降散可通过抑制NMDAR的磷酸化及提高GABAR mRNA表达水平来治疗大鼠神经病理性疼痛疾病,且对记忆力无明显影响。Objective: To investigate the effect of NMDAR and GABA receptors on neuropathic pain in rats treated with Shengjiang Powder and further investigate whether it may affect the memory by treatment with Shengjiang Powder. Methods: The rat model of chronic constriction injury(i CCI) was established. The rat model of chronic constriction injury of the sciatic nerve(i CCI)was established by using different doses of Shengjiang Powder. The rats in the i CCI model were treated by intragastric administration. P-NMDAR protein content and GABAR mRNA expression level were measured at different time points,and the rats were tested with water maze test at different time points. Results: The levels of p-NMDAR and GABAR mRNA in the model group were significantly lower than those in the normal group(P〈0.05). In the high dose group,the levels of p-NMDAR and GABAR mRNA level decreased significantly(P〈0.05). Compared with the model group,there was no significant effect on the number of wear platform. Conclusion: Shengjiang Powder can inhibit the NMDAR phosphorylation and increase the expression of GABAR mRNA in rats with neuropathic pain,and has no significant effect on memory.
