摘  要:为了研究无机焦磷酸盐(PPi)对17℃保存的猪稀释精液质量和精子顶体酶抑制剂(AI)水平的影响,试验选择2周岁长白猪精液分为4组,对照组精液保存液中不添加PPi,试验1组、试验2组、试验3组分别在精液保存液中添加10,50,100μmol/L的PPi,在17℃保存第1,2,3,4天采用精子质量分析仪(CASA)、考马斯亮蓝、低渗肿胀(HOST)法检测精子活率、顶体膜及质膜完整性,采用SDSPAGE及Western-blot法分析精子的AI水平。结果表明:保存2~4 d,试验2组、试验3组精子活率、顶体膜和质膜完整性均显著高于对照组和试验1组(P〈0. 05);保存3~4 d,试验1组、试验2组精子的AI水平显著高于对照组(P〈0. 05)。说明在精液保存液中添加PPi能显著改善17℃保存猪稀释精液的质量。In order to explore of inorganic pyrophosphate ( PPi ) on the quality and sperm acrosin inhibitor(AI) levels of diluted boar semen stored at 17 ℃, control group and treated groups were contained in this experiment, in which semen preservation with no PPi supplement in control group and, semen preservation with 10,50,100 μmol/L PPi in treated 1# group, treated 2# group and treated 3# group, respectively. All groups were stored at 17 ℃ for 1,2,3 and 4 days, and then sperm viability, acrosome and plasma membrane integrity were detected using sperm quality analyzer(CASA) ,eoomassie brilliant blue ,and low permeability swelling(HOST) methods respectively. SDS-PAGE and Western-blot methods were used to analyze the AI levels of semen from each group. The results showed that sperm viability, aerosome and plasma membrane integrity in treated 2# and 3# groups were signifieantly higher than those in control group and treated 1# group( P〈0. 05)on 2-4 days post treatment. The AI levels of semen in treated 1# and 2# groups were significantly higher than those in control group(P〈0. 05)on 3-4 days post treatment. The results suggested that PPi supplement can improve significantly the quality of diluted boar semen stored at 17 ℃.
