八种中草药乙醇提取物体外抗IBV增殖的研究

摘　　要：为了研究不同中草药的乙醇提取物对禽传染性支气管炎病毒（IBV）增殖的抑制效果,试验利用嵌合了荧光素酶的重组病毒IBV-3ab-luc与底物结合发出荧光的特性,定量检测了八种中药乙醇提取物在H1299细胞中的抗病毒活性,并对其中抑制效果最好的中药乙醇提取物的不同浓度药液对IBV增殖的抑制效果进行了检测。结果表明：皂荚乙醇提取物对IBV增殖的抑制效果最好,在病毒感染前期、中期加入皂荚乙醇提取物,病毒的数量与对照组相比均明显降低,抑制效力分别为4和5,且最佳使用浓度分别为0. 2 g/m L和0. 1 g/m L;白芍乙醇提取物在病毒感染中期加入效果较好,抑制效力为4;其他中草药乙醇提取物对病毒均无明显抑制效果。In order to study the inhibitory effect of ethanol extracts of different Chinese Herbal Medicines against avian infectious bronchitis virus （IBV） proliferation,the characteristics of recombinant luciferase-chimeric virus IBV-3ab-luc combining the substrate to produce fluorescence were used in this experiment for quantitatively detecting the antiviral activity of ethanol extracts of 8 kinds of Chinese Herbal Medicines in H1299 ceils and, for determining the inhibitory effect of different concentrations of the best ethanol extracts of Chinese Herbal Medicines on IBV proliferation. The results showed that ethanol extract of Gleditsia sinensis Lam had the best inhibitory effect on IBV proliferation and, a significantly lower virus number than that in control group when ethanol extract of Gleditsia sinensis Lain was added at the early and medium stages of viral infection. The magnitudes of the virus number ratio between control group and test group were 4 and 5 respectively and, the optimal concenrations were 0. 2 and 0. 1 g/mL respectively. Ethanol extract of Paeonia lactiflora had a good effect at the medium stage of viral infection and, the magnitude of the virus number ratio between control group and test group was 4. Other ethanol extracts of Chinese Herbal Medicines had no significantly inhibitory effect on the virus.
