摘 要:为探讨根际促生菌在种子引发中的作用,以吲哚乙酸(IAA)、赤霉素(GA3)和1-氨基环丙烷-1-羧酸脱氨酶(ACCD)为筛选指标,从扁穗雀麦(Bromus cartharticus,Vahl)幼苗根系中分离出可修复种子老化的植物根际促生菌BCR5,同时采用四因素四水平正交试验设计,分析了人工老化时间(A,24 h、48 h、72 h、96 h)、菌浓度(B,1×10^6cfu/mL、1×10^7cfu/mL、1×10^8cfu/mL、1×10^9cfu/mL)、种子引发方法(C,H2O引发、MgSO_4引发、H2O2引发、不引发)和引发时间(D,6 h、12 h、18 h、24 h)对扁穗雀麦老化种子萌发和幼苗生长的影响。结果表明:BCR5结合其他种子引发方法混合处理可降低丙二醛(MDA)含量,一定程度上恢复种子活力,提高老化种子的发芽率,并促进胚根和胚芽的伸长;菌浓度(B)是影响老化种子萌发和胚根伸长的主要因素,引发时间(D)主要影响胚芽生长;用菌株BCR5处理扁穗雀麦老化种子的最优方法为人工老化时间72 h、菌浓度1×107cfu/mL,结合H_2O_2引发处理18 h。此外,试验结果还表明适宜的人工老化处理可打破扁穗雀麦种子休眠。In order to explore the role of rhizesphere growth promoting bacteria in seed priming,a plant growth-promoting rhizobacteria(PGPR) BCR5 which can repair the seed aging was isolated from seedling root of rescue grass by evaluating the content of IAA,GA3 and ACCD. Meanwhile,an orthogonal experiment design method with four factors(the duration of artificial ageing,A; the bacterial concentration,B; different seed priming treatments,C; and priming time,D) and four levels(for factor A,the four level were 24 h,48 h,72 h and 96 h,respectively; for factor B,the four level were 1×106 cfu/mL,1×107 cfu/mL,1×108 cfu/mL and 1×109 cfu/mL,respectively; for factor C,the four level were water initiating,MgSO4 initiating,H2O2 initiating and none initiating,respectively; for factor D,the four level were 6 h,12 h,18 h and 24 h,respectively) was used to analyse the effect of BCR5 on seed germination and seedling growth of seed aging in rescue grass. The results showed that BCR5 mixed with other initiating methods could recover seed vigor,improve seed germination and increase seedling growth. Bacterial concentration(factor B) was the most important factor which influenced seed germination and seedling growth among the four factors,and priming time(factor D) mainly affected the cereal germ growth. The optimal method of repair the seed aging was artificial ageing with 72 h,bacterial concentration of 1×107 cfu/mL,H2 O2 priming and a priming time of 18 h.
