摘 要:为了研究MYH10基因在不同鸡种不同组织器官间的表达差异,试验利用定量PCR方法检测了MYH10基因在白羽肉鸡、大恒699肉鸡、藏鸡不同组织器官(胸肌、腿肌、下丘脑、垂体)、不同生长阶段(2周龄、6周龄、10周龄)的表达量。结果表明:在2周龄,白羽肉鸡公鸡胸肌中MYH10基因的表达量显著高于大恒699肉鸡(P〈0. 05),白羽肉鸡母鸡胸肌中MYH10基因的表达量极显著低于其他鸡种(P〈0. 01),藏鸡公母鸡腿肌中MYH10基因的表达量极显著高于其他鸡种(P〈0. 01);在6周龄,白羽肉鸡公鸡胸肌中MYH10基因的表达量显著高于大恒699肉鸡(P〈0. 05),藏鸡母鸡胸肌和腿肌中MYH10基因的表达量分别显著高于大恒699肉鸡和白羽肉鸡;在10周龄,藏鸡公母鸡胸肌中MYH10基因表达量极显著高于其他鸡种(P 〈0. 01)。说明MYH10基因参与了鸡肌肉组织的生长和发育过程的调控。The aim of the present study was to investigate the expression difference of MYH10 gene in different chicken breeds and tissues. The mRNA expression of MYH10 gene in different tissues(breast muscle,leg muscle,hypothalamus and hypophysis) and in different ages(2,6 and 10 weeks old) of Daheng chicken,White broiler and Tibetan chicken were detected by real-time quantitative PCR. The results showed that at the age of 2 weeks,the expression level of MYH10 gene in breast muscle tissue of White broilers was significantly higher than that of Daheng chickens(P〈0. 05). The expression level of MYH10 gene in breast muscle of female White broiler was significantly lower than that of other chicken breeds(P〈0. 01). The expression level of MYH10 gene in the muscle tissue of Tibetan chicken leg was significantly higher than that in other chicken breeds(P〈0. 01). At the age of 6 weeks,the expression level of MYH10 gene in breast muscle of white feather broiler was significantly higher than that of Daheng chicken(P〈0. 05),and the expression level of MYH10 gene in the breast and leg muscle of Tibetan chicken was significantly higher than that of Daheng chicken and White broiler,respectively. At the age of 10 weeks,the expression level of MYH10 in Tibetan chicken was significantly higher than that of other chicken breeds(P〈0. 01). The results indicated that MYH10 gene was involved in the regulation of the growth and development of chicken muscle tissue.
