摘  要:目的研究原位杂交技术(insituhybridization,ISH)在小鼠肠上皮具有潜在功能的目标基因筛选中的应用。方法应用原位杂交技术对小鼠小肠和结肠冰冻切片标本分别用标记的Clca4、Esam、Clca1、Rgmb探针进行检测。结果原位杂交技术检测出Clca4与Esam基因主要表达在小肠中,Clca1基因在小肠中的表达量高于结肠,而Rgmb基因在小肠和结肠中均高表达。结论原位杂交技术可以检测出不同基因在肠上皮的表达情况,该技术可以成为基因表达的定性与半定量研究手段,为肠上皮潜在功能基因的研究提供重要的筛选依据。Objective: To study the application of in situ hybridizaiton (ISH) in the screening of target genes with potential function in intestinal epithelium of mice. Methods: In situ hybridization was used to detect frozen sections of small intestine and colon using labeled Clca4, Clcal, Esam and Rgmb probes. Results: The expression of Clca4 and Esam genes in the small intestine was detected by ISH. The expression of Clcal gene in the small intestine was higher than that in the colon. The Rgmb gene was highly expressed both in the small intestine and colon. Conclusion: ISH can detect the expression of different genes in intestinal epithelium. This technique can be a qualitative and semi-quantitative research method for gene expression, and provides an important screening basis for the study of potential functional genes in intestinal epithelium.
