摘  要:利用正交试验,对广西藤县粉葛的外植体消毒、初代培养、继代培养以及生根培养进行了研究,总结出一套适合广西藤县粉葛组培苗生产的繁育技术。结果表明:外植体采用75%酒精浸泡15 s,再用0.1%HgCl2消毒7.2 min,便能取得较好的效果,存活率超过50%;无菌外植体接种到MS+6-BA 0.2~0.3 mg/L+NAA 0.1~0.5 mg/L培养基上进行初代培养,腋芽萌发率达75%以上;初代培养诱导的腋芽再用MS+6-BA 0.3 mg/L+NAA 0.05 mg/L进行继代培养,其繁殖系数能达到4以上;最后,以1/2MS或1/4MS加NAA 0.01~0.05 mg/L培养基进行生根培养,其生根效果最好。研究结果将为广西粉葛健康种苗生产提供技术支撑。This paper studied disinfecting way, primary culture, subculture and rooting culture of Puerariathomsonii Benth in Tengxian Guangxi by orthogonal test, and summaried a set of industrialized seedling technology.The results showed that, the survival rate of explant could exceed 50% after disinfection for 15 sec with 75%Ethanol and 7.2 min with 0.1% HgCl2. The axillary buds induction rate were more than 75% after the asepticexplant was inoculated into MS+6-BA 0.2~0.3 mg/L+NAA 0.1~0.5 mg/L. Then these axillary buds were transferredto MS+6-BA 0.3 mg/L+NAA 0.05 mg/L for subculture, the propagation coefficient was more than 4 times. The mostoptimal rooting medium was 1/2 MS or 1/4MS supplemented with NAA 0.01~0.05 mg/L. The results will providetechnical support for seedling production of Pueraria thomsonii Benth in Guangxi.
