摘 要:利用显微照相和DNA提取等技术,研究了外源紫杉醇作用下悬浮培养南方红豆杉细胞 (Taxus chinensis var. Mairei) 的凋亡和对紫杉醇(taxol)的耐受性。利用荧光显微照相技术观察到,外源紫杉醇作用下,随培养时间延长, 南方红豆杉细胞发生明显核分化;应用DNA电泳观察到细胞DNA断裂形成典型的180bp及其整数倍梯形条带(DNA ladders), 应用TUNEL检测发现DNA的3,-OH断端可被特异性标记, 证实了加入紫杉醇后南方红豆杉细胞发生了凋亡。研究结果说明南方红豆杉细胞对紫杉醇的耐受性是有限度的, 并证明了植物细胞有类似于动物细胞的凋亡特征, 也表明该体系可以作为研究植物细胞凋亡的实验模型。Apoptosis and the tolerance of Taxus chinensis var. mairei cells in suspension cultures treated with taxol were studied by microscopy and total DNA agarose gel electrophoresis. By using fluorescence microscopy, it was observed that the cell differentiation appeared after the addition of taxol. DNA extracted from the treated cells formed a characteristic 搇adders?on agarose gel electrophoresis. TdT-mediated dUTP nick end labeling (TUNEL) analysis of the cells revealed that the nuclear DNA strand breaks could be identified by labeling free 3-OH termini. The results show that taxol causes apoptosis of Taxus chinensis var. mairei cells and Taxus chinensis var. mairei cells are able to tolerate to taxol at the low concentration. The characteristics of apoptosis in plant cells are very similar to that in animal cells. Taxus chinensis var. mairei cells can be taken as a model for studying apoptosis.
