Effect of Copper on Proliferation and Autocrine of IGF-Ⅰ,IGFBP_3 in the Young Pig Chondrocytes in Vitro

摘  要:体外分离、培养仔猪关节软骨细胞 ,然后在细胞培养液中分别添加 0、7.8、15 .6、31.2、6 2 .5 μm ol/ L 铜。结果表明 ,软骨细胞在 4种浓度的铜中可存活并增殖 ,而且能增加胰岛素样生长因子 (IGF- )、胰岛素样生长因子结合蛋白(IGFBP3 )的分泌量。但随铜浓度的增加 ,其存活率、增殖率、3 H- Td R掺入率及 IGF- 、IGFBP3 的分泌量有明显的差异。且以培养液中添加 31.2μmol/ L铜对软骨细胞的增殖作用最强 ,增殖率、3 H- Td R掺入数、IGF- 、IGFBP3 的分泌量显著高于对照组 (P <0 .0 1)。表明 31.2 μmol/ L 铜浓度是促进体外软骨细胞增殖和自分泌 IGF- 、IGFBP3 的最适浓度。Chondrocytes from the lateral trochler ridge of the distal femur in one-day pig were cultured in medium supplemented with 0,7.8,15.6,31.2,62.5 μmol/L copper. IGF-1 and IGFBP 3 levels in culture medium were determined with radioimmunoassay (RIA). DNA synthesis in chondrocytes was measured by 3H-thymindine incorporation. The results were as follows: Proliferation-promoting activity and incorporation of 3H-thymindine in chondrocytes increased in culture medium added with 7.8,15.6, 31.2 and 62.5 μmol/L Cu. The content of IGF-Ⅰand IGFBP 3 in chondrocytes was also enhanced significantly in culture medium with 15.6,31.2,62.5 μmol/L Cu. The results showed that effects of copper supplemented in culture medium on chondrocyte proliferation were related to the increasing levels of IGF-1 and IGFBP 3 in culture medium which were induced by copper.
