摘  要:用无血清培养系统研究了牛腔前卵泡的体外培养。直径为 10 0~ 2 0 0μm的牛腔前卵泡在添加 L -谷氨酰胺、BSA、睾酮、转铁蛋白和硒的 Mc Coy′s5 a培养液中培养 ,卵泡保持正常的形态结构并持续生长 ,培养 10 d左右形成卵泡腔 ,成腔率约 5 0 %。培养液中添加胰岛素对卵泡直径的增长和卵泡腔的形成有明显的促进作用 ,但添加 FSH对腔前卵泡的生长未表现促进作用。A serum-free culture system was used to study the in vitro culture of the preantral follicles from bovine ovaries. Follicles with diameters of 100-200 μm, dissected mechanically from ovarian cortical slices, were cultured individually in 96-well plates with 250 μL McCoy′s 5a medium supplemented with 20 mmol/L Hepes, 3 mmol/L L-glutamine, 0.1% BSA, 29 μg/L testosterone, 2.5 mg/L transferrin and 4 μg/L selenium. The follicles were incubated in a humidified atmosphere with 5% CO 2 at 37℃. The culture medium was changed every 6 days. The results showed that the follicles grew continuously maintaining their normal morphology. Antrum formation occurred after about 10 days of culture. The addition of insulin to the culture medium stimulated the growth of preantral follicles. However, similar effect was not shown for the addition of FSH.
