摘  要:链脲佐菌素引起的糖尿病大鼠用胰岛素治疗6天后,治疗组(n=4)与未治疗组(n=20)比较:(1)体重和血清胰岛素浓度显著增加,而摄食量,饮水量和血糖均显著降低;(2)脂肪细胞膜胰岛素结合率显著降低,其胰岛素受体数目减少但受体亲和力没有改变;(3)基础(不加胰岛素时)和胰岛素刺激的脂肪细胞葡萄糖氧化均显著增加。提示胰岛素治疗虽使靶细胞上胰岛素受体数目减少,但可显著提高靶细胞对胰岛素的反应性,即可使受体后胰岛素抵抗减轻。The insulin binding to adipocyte membranes and glucose oxydation of adipose cells were measured in streptozotocin induced diabetic rats treated with insulin for 6 days. The results showed that 1) body weight and serum insulin were significantly increased (P<0.05~0.01), while food and water intake and blood glucose were significantly decreased (P<0.05~0.01) in insulin-treated diabetic rats (n = 14) as compared with untreated diabetic rats (n = 20); 2) adipocyte membranes from diabetic rats treated with insulin bound much less insulin than those from control diabetic rats with a decreased number of insulin receptors and a unchanged affinity for receptors of insulin; 3)a significantly increased glucose oxydation in adipocyte of insulin-treated diabetic rats was found as compared with those of control diabetic rats whether the data were analyzed based on the absolute values or the percentage increased above basal levels (in absence of exogenous insulin). The above data suggested that insulin therapy led to decreased number of insulin receptor but increased responsiveness of target cells to insulin, i.e., it might decrease insulin resistance at postreceptor sites in insulin-deficient diabetic rats.
