摘  要:报道了家蚕杆状病毒表达hGM CSF产业化过程中几个问题的研究结果。鲜茧在 4~ 7℃保存可显著抑制蚕蛹的发育 ,但随冷藏时间的延长 ,死笼率明显上升 ,接种成功率显著下降 ;接种蚕蛹血淋巴中hGM CSF的活性与鲜茧保存时间有明显的负相关关系 ;生产工艺不同 ,蚕蛹冻干粉中hGM CSF的活性有明显差异 ;60 Co辐照可使冻干蚕蛹粉中微生物的数量减少 ,但同时蚕蛹中hGM CSF的活性也随辐照剂量的增加而有所降低 ,hGM CSF蚕蛹粉经 30 0 0Gyγ射线辐照后 ,口服仍使小鼠白细胞明显上升。Our research focused on important problems related to industrialization of hGM CSF expressed in silkworm baculovirus system.Development of fresh silkworm pupa was clearly repressed when they were kept at 4 to 7℃ and their death rate greatly increased while the rate of successful inoculation significantly decreased with elongation of cryopreservation time.The activity of hGM CSF in hemolymph of inoculated pupa was negatively correlated with preservation time.And the activity of hGM CSF in freeze dried powder of pupa differed greatly because of different production crafts.Exposure to 60 Co radiation decreased significantly the amount of microbes in pupa powder,while the activity of hGM CSF in freeze dried powder of pupa decreased evidently at the same time.After hGM CSF pupa powder treated with 3 000 Gy γ radiation was orally administrated to mice,the amount of leukocytes in mouse clearly increased.
