摘 要:用烫伤和制动水浸应激大鼠观察肝细胞胰岛素受体的变化和地塞米松与水飞蓟宾的影响。烫伤大鼠血浆葡萄糖和胰岛素含量高于正常大鼠,出现创伤后胰岛素耐受。烫伤后72h大鼠肝细胞咦岛素受体最大结合容量(Ro)和最大结合率(Bo/T)有明显降低趋势,但亲和力( e. f)无明显变化。制动水浸应激大鼠受Ro和Bo/T均有明显降低趋势, e和 f显著升高。地塞米松有使烫伤后72h大鼠的Ro明显增加趋势, e明显降低;水飞蓟宾处理则使Ro和Bo/T均有显著增高趋势,亲和力( e. f)无明显变化。结果提示创伤后胰岛素耐受与胰岛素受体缺陷有关,水飞蓟宾对创伤后糖代谢紊乱可能起有益作用,而地塞米松则否。The purpose of this study is to examine the alterations in hepatocyte insulin receptors in rats subjected to scald and restrained water immersion stress and the effects of dexametha- s one and silybin.In scalded rats,plasma gluccse and insulin levels were higher than that of normal rats,suggesting the occurrence of insulin resistance following injury.The maximal binding capacity(Ro)and highest insulin binding rate(Bo/T)of hepatocyte insulin receptors were significantly reduced in rats 72h postscalding,while there was no obvious change in affinity( e f).The Ro and Bo/T of the restrained water immersion stress rats were also reduced significantly,but the affinity was increased.Dexamethasone signifificantly enhanced Ro in rats 72h postscalding with significant decrease in affinity( e).Silybin significantly enhanc- ed Ro and Bo/T 72h postscalding with no obvious change in affinity.It is suggested that in- sulin resistance following injury may be a censecuence of receptor defect of the target cells, and for disturbance of glucose metablism fellowing injury,it is not beneficial to use dexame- thasone,but silybin may be used with benefit
