摘  要:研究利用 β NADPH d组织化学染色法检测一氧化氮合酶 (Nitricoxidesynthase,NOS)在正常肉鸡、亚临床腹水鸡和腹水鸡肺组织中的分布和活性变化。结果显示 :在正常肉鸡肺组织中 ,气道上皮和肺大、中血管内皮出现NOS阳性染色 ,而外径小于 10 0 μm的小血管却很少见阳性染色 ;腹水鸡肺内大、中血管内皮着色较正常肉鸡深 ,血管平滑肌也有阳性反应 ,在正常肉鸡不出现阳性染色的肺小血管出现了强阳性染色 ,肺小血管壁增厚 ,无肌性血管肌化 ,染色很深 ;亚临床腹水鸡有部分肺小血管内皮或平滑肌着色。研究结果表明正常肉鸡肺血管内皮有NOS活性 ,说明由NOS催化生成的NO对正常肉鸡肺血管张力有一定调节作用 ;腹水鸡肺小血管平滑肌层和内皮NOS表达增强 ,这可能与肺血管中膜增厚有关。By using β-NADPH-diaphorase histochemical staining techniques,the localization and activity of nitric oxide synthase in the lungs of normal,preascites and ascites broilers were investigated in this paper.In the normal broilers,NADPH-d was distributed in the epithelium of air pathway and endothelium of large and medium (ID>100 μm)vessels but was not detected in the small vessels (ID<100 μm),and there was an absence of NADPH-d staining in the smooth muscle layer of all pulmonary vessels.However,in ascites broilers,NADPH-d staining in the endothelium of large and medicum pulmonary vessels increased,the smooth muscle layers also showed positive staining,NADPH-d became markedly positive in the endothelium and smooth muscle layer of small vesseles with thicker walls and of muscularized nomuscular artery.Some endothelium and smooth muscle layer of small pulmonary vessels of preascites broilers showed positive staining.These results suggest that the NO induced by the NOS might regulate vasodilation of pulmonary vessels of normal broilers and the increased expression of NOS in small pulmonary vessels of ascites broilers might be associated with the chicken of medial smooth muscular layer of pulmonary vessels.
