“金龙胶囊”抑制HL-60细胞生长并诱导细胞凋亡

摘　　要：目的:研究中药金龙胶囊冻干粉(Jin Long Capsules,简称JLC)对人白血病细胞的作用及其机制。方法:应用台盼兰拒染法观察0.01mg/ml、0.02mg/ml、0.04mg/ml JLC对HL-60细胞的抑制作用,光镜及透射电镜观察细胞结构变化,DNA电泳、流式细胞术等检测细胞凋亡。结果:1.JLC可抑制HL-60细胞的生长,这种作用呈剂量和时间依赖性;2.JLC作用后细胞出现典型的凋亡形态学改变,DNA片段化,流式细胞仪可检出亚G_1期细胞;3.JLC作用后细胞阻滞于G_0/G_1期,S期细胞减少。结论:金龙胶囊冻干粉对白血病细胞有诱导凋亡作用,主要作用于S期。Objective To study the mechanism of Jin Long Capsules (JLC) on human leukemia cell inhibition. Methods HL - 60 cells were treated with JLC at different concentrations. The growth inhibition was analyzed by the method of trypan blue excluding staining, cell apoptosis by light microscopy, transmission election microscopy, flow cytometry and agarose gel electrophoresis. Results ①Treatment of HL-60 cells with JLC remarkably inhibited the cell growth. ②Apoptosis of HL-60 cells could be induced by JLC at concentration of 0.01mg/ml or higher. JLC induced apoptosis of HL-60 cells in a dose - and time - dependent manner.③Apoptotic cells increased with Go/Gj phase cells increasing and S phase cells decreasing. Conclusion JLC could induce apoptosis of HL-60 cells, especially the S phase cells.
