摘  要:比较了超声提取与回流提取法在提取牛膝中活性成分蜕皮甾酮的提取效率 ,在频率为 110 0 k Hz时超声提取 4 0 min可获得稳定的提取效率。采用 Shim- Pack CL C- ODS色谱柱 ,甲醇∶水 =4 0∶ 6 0 (V/ V,% )为流动相 ,在波长 2 4 8nm下 ,测定牛膝中蜕皮甾酮 ,线性范围为 2— 2 0 0 μg/ m L(r=0 .9998) ,检出限为0 .0 5 2μg/ m L (S/ n=3) ,加标回收率为 98.3%— 10 3%。A sensitive and precise RP HPLC method for the determination of ecdysterone in achyranthes bidentata blume was developed.The ecdysterone was extracted with ultrasonic extraction method.The ecdysterone was separated on Shim Pack CLC ODS column and determined at 248nm by using the solvent system methanol water(40∶60)with flow rate of 1.0 mL/min .There is a good linear relationship within the range of 2-200μg/mL( r =0.9998)of ecdysterone and the recovery is 98.3%-103% .The proposed method is simple,sensitive and effective for the determination of the ecdysterone in achyranthes bidentata blume from different locations.
