摘  要:【目的】探讨大鼠前额叶皮层神经元N 甲基 D 天冬氨酸 (NMDA)受体通道的特性 ,为研究人脑前额叶锥体神经元的NMDA通道提供动物实验依据。【方法】对急性分离的大鼠前额叶的神经元进行细胞贴附式单通道膜片钳记录。【结果】前额叶神经元NMDA受体通道的开放呈现多电导状态 ,表现为 5 5 1,36 5 ,18 6 pS 3种状态 ,以 18 6 pS为优势电导。开放时间主要表现为快时程和慢时程两种状态 ,时间常数均值分别为 :(0 5 5± 0 38)ms和 (13 5 9± 3 2 6 )ms,以短时程为主。关闭时间常数为 (0 34± 0 2 7)ms和 (19 73± 2 4 1)ms。开放概率为 (0 2 5± 0 0 9)。【结论】前额叶神经元NMDA受体通道呈多电导 ,以低电导 18 6pS为优势。开放时程短者占优势 ,关闭时间短暂 ,开放概率高 ,表明前额叶锥体神经元NMDA受体通道的活动以快速短暂样开放为主。To investigate the single channel properties of N methyl D aspartate(NMDA) receptors in prefrontal cortex neurons of the rats Single channel currents were recorded from newly dissociated pre frontal cortex neurons with cell attached configuration of patch clamp Three conductances were observed from the frontal cortex neurons, which are referred as large, medium, and small conductance channels The mean single conductances were 55 1, 36 5, 18 6 pS respectively, with 18 6 pS as the predominant one The mean open time histograms were adequately fitted with two exponentials The mean open time constant were (0 55±0 38) ms and (13 59±3 26) ms. The open probability was (0 25±0 09).[Conclusion] The low conductance and short openings are predominant in prefrontal cortex, but the opening probability is high.
